Digital holographic microscopy (DHM) offers label-free, full-field imaging of live-cell samples by capturing optical path differences to produce quantitative phase images. Accurate cell segmentation from phase images is crucial for long-term quantitative analysis. However, complicated cellular states (e.g., cell adhesion, proliferation, and apoptosis) and imaging conditions (e.g., noise and magnification) pose significant challenge to the accuracy of cell segmentation. Here, we introduce DL-CSPF, a deep-learning-based cell segmentation method with a physical framework designed for high-precision live-cell analysis. DL-CSPF utilizes two neural networks for foreground-background segmentation and cell detection, generating foreground edges and "seed points." These features serve as input for a marker-controlled watershed algorithm to segment cells. By focusing on foreground edges and "seed points", which have lower information entropy than complete cell contours, DL-CSPF achieves accurate segmentation with a reduced dataset and without manual parameter tuning. We validated the feasibility and generalization of DL-CSPF using various open-source and DHM-collected datasets, including HeLa, pollen, and COS-7 cells. Long-term live-cell imaging results further demonstrate that DL-CSPF reliably characterized and quantitatively analyzed the morphological metrics across the cellular lifecycle, rendering it a promising tool for biomedical research.

This study investigates the synthesis of aromatic nitriles using an evolved variant of OxdF1 (L318F/F306Y), an aldoxime dehydratase from Pseudomonas putida F1, engineered for improved catalytic efficiency toward benzaldehyde oxime. The double OxdF1 (L318F/F306Y) mutant effectively catalyzes the conversion of various benzaldoxime derivatives to the corresponding nitriles. Due to the enzyme's inherent instability, immobilized whole-cell systems are employed in a flow reactor to improve its stability and broaden its applicability, with the biotransformation of benzaldehyde oxime and 2,6-difluorobenzaldehyde oxime serving as case studies.

Objective: To assess the production of nitric oxide and endothelin in off-pump coronary artery bypass grafting by comparing two techniques of internal thoracic artery preparation: skeletonized and pedicled without endothoracic fascia.
Methods: In this prospective, randomized clinical study, 40 patients undergoing off-pump coronary artery bypass grafting were randomized according to internal thoracic artery preparation technique into the skeletonized or pedicled (without endothoracic fascia) groups (n=20 each). Endothelial expression of CD31 was evaluated by means of immunohistochemistry and en-face immunofluorescence.

In cancer research, multiplexed imaging allows detailed characterization of the tumor microenvironment (TME) and its link to patient prognosis. The integrated immunoprofiling of large adaptive cancer patient cohorts (IMMUcan) consortium collects multi-modal imaging data from thousands of patients with cancer to perform broad molecular and cellular spatial profiling. Here, we describe and compare two workflows for multiplexed immunofluorescence (mIF) and imaging mass cytometry (IMC) developed within IMMUcan to enable the generation of standardized data for cancer tissue analysis.
