Article Abstract

A simple and highly chemoselective heteroannulation protocol for the synthesis of polysubstituted heterocycles is reported. In this work, various oxa-aza[3.3.3]propellanes were synthesized via a sequential one-pot reaction of ninhydrin, malononitrile and nitroketene acetal or nitroketene aminals in water at ambient temperature. This successive Knoevenagel condensation/Michael addition/cyclization sequence features high atom economy, excellent efficiency, and the use of water as a solvent without the need for metal catalysts or external activators. This method can be presented as a good example of group-assisted purification (GAP), in which traditional purification techniques such as recrystallization and column chromatography are avoided and pure products are obtained simply by washing the crude products.

We herein describe a novel decarboxylation/cyclization sequence involving a three-component reaction of dialkyl 2-(alkoxymethylene)malonate, amines, and terminal alkyne ester or internal alkyne ester catalyzed by CsCO under microwave conditions. These two types of highly chemo- and regioselective transformations were accomplished by different reaction channels to furnish a wide range of functionalized 8-hydroxyisoquinoline-1(2)-ones (21 examples) and 2-pyridones (18 examples) in good to excellent yields and might provide new opportunities for the discovery of N-heterocyclic drugs and other functional molecules.

Glioblastoma (GBM) is a highly malignant brain tumor with limited treatment options and poor prognosis. GBM exhibits resistance to conventional therapies, including temozolomide (TMZ), radiotherapy, and immunotherapy, partly due to immunosuppressive mechanisms such as programmed death-ligand 1 (PD-L1) overexpression. Catalytic reduction of quinolines has gained continuous interest in both academia and industry, providing direct and efficient access to tetrahydroquinolines or 1,2-dihydroquinolines. The catalytic preparation of tetrahydroquinolines has been extensively studied by transition metal complexes. By contrast, the related catalytic synthesis of 1,2-dihydroquinolines remains underdeveloped due to the difficulties in achieving precise control over both chemo- and regioselectivity.
