As supramolecular assemblies, polypseudorotaxanes (PPR) exhibit inherent advantages in modular adaptability and structural programmability, with the potential to build tuneable platforms integrating various functionalities. Here we report the "one-pot" preparation of a self-assembled thiol-rich PPR (SPPR), where thiolated-α-cyclodextrins (SHαCD) spontaneously thread onto polymers, and are then crosslinked into a three-dimensional network by the thermally-triggered oxidation of thiols into disulfide bonds. The dynamic thiol groups along the SPPR provide remarkable modularity for the functionalization of thiophilic metal nanoparticles (NPs), exemplified by two application vectors.

Upscaling, toxicity and efficacy of multifaceted dressing embedded with dsirna-loaded gold nanoparticles for enhancing diabetic wound treatment.

Poor vascularization and infections hinder diabetic wound healing, posing challenges in therapy development. A multi-action approach incorporating Dicer-substrate small interfering RNA (DsiRNA) against the prostaglandin transporter (PGT) gene and gold nanoparticles (AuNPs) into a Pluronic F-127 (PF127) gel was developed. This study aimed to upscale AuNP biosynthesis using Lignosus rhinocerotis (tiger milk mushroom, TMM) extract and chitosan as stabilizers.

Photodimerization-Mediated Intramolecular Cross-Linking of Thermoresponsive Dendronized Cyclodextrin-Based Polyrotaxanes.

International Joint Laboratory of Biomimetic and Smart Polymers, School of Materials Science and Engineering, Shanghai University, Shanghai, China.

Intramolecular cross-linking of mechanically interlocked polyrotaxanes (PRXs) is promising to create novel single-chain nanoparticles with distinct architectures and properties. Here, we report intramolecular cross-linking of thermoresponsive dendronized cyclodextrin-based PRXs through photodimerization mediated by dendritic confinement. These PRXs are based on polyethylene glycol axle, which are dendronized with cinnamate-cored threefold dendritic oligo(ethylene glycol)s (OEGs) to contribute dually thermoresponsive and photo-cross-linkable functions.

Diabetes-related foot ulcers (DRFUs), affecting one-third of individuals with diabetes, are a leading cause of hospitalization. Current treatment options to expedite healing are limited, with the effectiveness of potential therapeutics reduced by the hostile environment inherent to most diabetes-related wounds. Topical delivery vehicles that protect drugs from the protease-enriched wound environment, whilst displaying a slow-release profile, are needed to facilitate wound closure.

Superstretchable and Tough Physical Cross-Linking Hydrogels with Dual pH and Temperature Responsiveness for In Vivo Treatment of -Infected Wounds.

Biomacromolecules

September 2025

Key Laboratory of Product Packaging and Logistics, Packaging Engineering Institute, College of Packaging Engineering, Jinan University, Qianshan Road 206, Zhuhai 519070, China.

Developing a single hydrogel with comprehensive wound healing functions is challenging. We developed a composite hydrogel wound dressing with thermal and pH responsiveness, demonstrating antibacterial and biocompatible properties in rats with -infected wounds. Sodium alginate cross-linked by Ca served as the continuous phase, while thermosensitive poly(isopropylacrylamide), laponite, lecithin, gelatin, and antibacterial ZIF-8 supported by tannic acid-induced Ag nanoparticles formed the dispersed phase.
