Parkinson's disease (PD) is a common neurodegenerative motor disorder and is frequently accompanied by several non-motor symptoms. Among the latter, gastrointestinal problems, including constipation, often emerge years before the manifestation of locomotion difficulties and before a diagnosis of PD is established. A recent study proposed a method to measure constipation in a PD fly model that expresses wild-type human alpha-synuclein. In this study, we examined the feasibility of this method and tested whether other genetic fly PD models equally result in constipation. Available fly lines were used to measure the constipation level in autosomal dominant and recessive PD models, including three different mutant human alpha-synuclein-expressing Drosophila lines and Pink1- and Parkin-deficient flies. These flies and their corresponding controls were placed on blue-colored food one day before the experiment to induce blue fecal spots. On the day of the experiment, these flies were transferred to standard cornmeal. The percentage of blue fecal spots to total fecal spots was recorded and calculated every 90 min. The different fly PD models show slower or incomplete expulsion of blue-colored food compared to their respective control flies. Hence, a previously proposed method to measure constipation was validated and applied to various fly models of different forms of genetic PD.

Data were collected between April 2023 and April 2024 at the Adult Detoxification Center of a psychiatric hospital in Istanbul, Türkiye. The sample consisted of a total of 261 individuals with substance use disorders (218 males and 43 females) who met the inclusion criteria.

Ethnopharmacological Relevance: The Shi Pi Zeng Ye Formula (SPZY), a traditional Chinese herbal compound, is empirically used for qi and yin replenishment and has been prescribed for managing functional constipation (FC) comorbid with depression. Although its clinical efficacy is recognized, the active constituents and their precise mechanisms of action in treating FC comorbid with depression have yet to be fully determined.
Aim Of The Study: This research aims to elucidate the efficacy and mechanisms underlying the therapeutic effects of SPZY on FC comorbid with depression, employing a single-arm study design alongside mass spectrometry, network pharmacology, and molecular docking.

Despite advancements in surgical techniques and improved survival rates for anorectal malformation (ARM) patients, the lack of a standardized way for assessing bowel function and quality of life remains a challenge. Variability in scoring outcomes exists between systems due to inconsistencies in criteria and precise definitions, such as soiling, constipation, and continence. While older tools lack proper validation, newer scales, including the Rintala and Baylor Continence Scale (BCS), offer more structured assessment.

Background: Autonomic symptoms are among the most important factors determining the quality of life in patients with Parkinson's disease (PD). This study aimed to assess the profile of autonomic dysfunction symptoms in three groups of patients with genetic PD, carrying mutations in , , and genes, compared with subjects with sporadic PD.
Methods: This case-control observational secondary analysis of prospectively collected data was performed on 742 patients (485 in the sporadic group, 165 in the LRRK2 group, 85 in the GBA group, and nine in the PRKN group).
