Article Abstract

The roles of the alternative oxidase (AOX) pathway in plant physiology and metabolism are of increasing interest. AOX was found to regulate the growth of Enhalus acoroides seedlings, but its specific mechanism and physiological significance are unclear. In this study, the roles of the AOX pathway during E. acoroides seedling establishment were clarified by investigating the relationships between metabolism and the AOX pathway at the physiological and molecular levels. Results showed that inhibiting the AOX pathway causes the accumulation of reducing equivalents, and further results in the inactivation of the PSII reaction center and destruction of the PSII electron receptor side in E. acoroides seedlings, which decreased photosynthetic activity and increased HO content. Meanwhile, the accumulation of reducing equivalents also restricted mitochondrial respiratory metabolism (including glycolysis, the tricarboxylic acid cycle, the pentose phosphate pathway and oxidative phosphorylation). In addition, when the AOX pathway was inhibited, the gene expressions related to photosynthesis and respiratory metabolism were generally down-regulated. The above results indicate that inhibiting the AOX pathway affected metabolism through disturbing photosynthetic and respiratory metabolic processes, resulting in an inability to satisfy the material (saccharides, proteins, lipids, and nucleotides) and energy requirements of various physiological processes, thus stunting the growth of seagrass seedlings. This study reveals that the AOX pathway accelerates the production of intermediate metabolites in key metabolic pathways through energy redistribution in seagrass, which has a very positive significance for the establishment of seagrass seedlings.

Unlabelled: The mevalonate pathway produces sterols and isoprenoids that support cancer cell growth, yet its broader metabolic functions remain incompletely defined. Here, we show that this pathway sustains amino acid biosynthesis by promoting mitochondrial NAD⁺ regeneration through ubiquinone-dependent electron transport. Statin-mediated inhibition of the mevalonate pathway impairs oxidative phosphorylation, lowers the NAD⁺/NADH ratio, and suppresses de novo serine and aspartate synthesis, thereby activating the GCN2-eIF2α-ATF4 amino acid deprivation response.

Hormones such as abscisic acid and brassinosteroids ameliorate stress tolerance in plants. The present investigation demonstrates the importance of brassinolide, an active form of brassinosteroids, in sustaining photosynthesis under high light (HL). The addition of brassinolide to mesophyll protoplasts activates the mitochondrial electron transport chain (mETC), particularly through an alternative oxidase (AOX) pathway.

Salt stress severely hampers plant growth and crop productivity. Defects in Golgi α1,2-mannosidase I MNS1 and MNS2 proteins, essential for N-glycan maturation, lead to severe root growth inhibition and swollen tips in Arabidopsis under salt stress. Here, we reported sgam1, a suppressor of mns1 mns2, exhibiting threshold-dependent suppression of salt sensitivity.

Unlabelled: Acute salinity stress critically impacts aquaculture efficiency by inducing physiological disruptions in fish. This study investigated the molecular adaptation mechanisms of exposed to 10 ppt NaCl for 96 h, integrating transcriptomic and metabolomic analyses of gill and kidney tissues. In transcriptomics, 2,554 and 1,066 differentially expressed genes (DEGs) were identified in gills and kidneys, respectively, with significant enrichment in pathways related to energy metabolism (glycolysis, oxidative phosphorylation), membrane dynamics (glycerophospholipid metabolism), and immune-osmoregulatory crosstalk (HIF-1, TNF, and Jak-STAT signaling).

Fruit respiration varies throughout development, reflecting dynamic changes in energy demand and metabolic activity. Grapes grow more rapidly during the preveraison stage, as cell division and expansion drive berry enlargement before the onset of ripening. The plant mitochondrial electron transport chain contains two terminal oxidases; alternative oxidase (AOX) and cytochrome c oxidase (COX), the activities of which are central to predicting patterns of growth in plant organs.
