The manchette is a transient skirt-like structure consisting of microtubules (MTs) and filamentous actin (F-actin) surrounding the elongating sperm head during spermiogenesis. It is pivotal in sperm head shaping controlled by the acrosome-acroplaxome-manchette complex, acrosome formation, and flagellar assembly by microtubular-based protein delivery. Defects in the manchette frequently lead to teratozoospermia concomitant with oligozoospermia and asthenozoospermia, but the pathogenic mechanism underlying manchette function and its role in male infertility remain poorly understood. In this review, we systematically described the assembly and disassembly of the manchette, intra-manchette transport (IMT) and its regulatory model, the function and mechanism of manchette and IMT in regulating sperm head shaping and flagellar assembly during spermatogenesis; summarized the research progress of manchette-related genes related to male infertility; and listed the manchette-related proteins in knockout mouse models and clinical cases, which provide the theoretical basis for an in-depth understanding of the molecular mechanism of manchette involved in spermatogenesis and male fertility for understanding the potentially developing treatments for infertility and reproductive disorders.

This study evaluated the cryoresistance of stallion sperm frozen by ultra-rapid (UR) methods using microspheres and straws or by the conventionally-slow (CS) method. Sixteen ejaculates from four stallions were each divided into three aliquots according to the freezing method: UR freezing in 30-μL spheres (UR-Spheres) by direct immersion in liquid nitrogen (LN); UR freezing in 0.25-mL straws (UR-Straws) by direct horizontal submersion in LN; and CS freezing in LN vapors.

Characterising body and reproductive morphometry and their association with epididymal sperm quality can contribute to the conservation of sambar deer (Rusa unicolor). Five adult males maintained in captivity at the Getúlio Vargas Zoobotanical Park (Salvador, BA, Brazil) were captured, anaesthetised, and subjected to bilateral orchiectomy as part of a population-control strategy. Body measurements included head circumference, thoracic diameter, total length, withers height, and body weight.

In the evaluation of male infertility, precise assessment of sperm functional competence has surpassed the requirements of conventional semen parameters. Existing computer-aided analysis systems are deficient at the molecular diagnostic level and also face challenges in live-cell fluorescence quantification. To address these issues, we have developed a novel integrated computational-imaging platform that combines a fine-tuned You Only Look Once version 8 (YOLOv8) architecture, tailored for the EVISEN dataset, with dual-probe fluorescence microscopy image segmentation, enabling simultaneous quantification of intracellular pH (pHi) and mitochondrial DNA G-quadruplexes (mtDNA G4s).

In mammals, sperm formation is completed in the seminiferous tubules within the testis, and sperm maturation occurs during the epididymal transit of the spermatozoa. Sperm morphology drastically changes when abnormal spermatozoa migrate from the testis to the epididymis. Detailed molecular mechanisms for sperm survival in the epididymis have not been determined yet.

Cryopreservation of ram sperm is a cornerstone in reproductive biotechnology but is often accompanied by oxidative damage that compromises post-thaw sperm quality. Curcumin, a natural polyphenol with potent antioxidant properties, may offer protection against cryo-induced injuries. This study evaluated the effects of curcumin supplementation at two concentrations (10% and 20%) in semen extenders on the functional and molecular characteristics of ram spermatozoa.
