Motivation: Drug side effects refer to harmful or adverse reactions that occur during drug use, unrelated to the therapeutic purpose. A core issue in drug side effect prediction is determining the frequency of these drug side effects in the population, which can guide patient medication use and drug development. Many computational methods have been developed to predict the frequency of drug side effects as an alternative to clinical trials. However, existing methods typically build regression models on five frequency classes of drug side effects and tend to overfit the training set, leading to boundary handling issues and the risk of overfitting.
Results: To address this problem, we develop a multi-source similarity fusion-based model, named multi-source similarity fusion (MSSF), for predicting five frequency classes of drug side effects. Compared to existing methods, our model utilizes the multi-source feature fusion module and the self-attention mechanism to explore the relationships between drugs and side effects deeply and employs Bayesian variational inference to more accurately predict the frequency classes of drug side effects. The experimental results indicate that MSSF consistently achieves superior performance compared to existing models across multiple evaluation settings, including cross-validation, cold-start experiments, and independent testing. The visual analysis and case studies further demonstrate MSSF's reliable feature extraction capability and promise in predicting the frequency classes of drug side effects.

The μ-opioid receptor (μOR) is the primary drug target of opioid analgesics such as morphine and fentanyl. Activation of μORs in the central nervous system inhibits ascending pain signaling to the cortex, thereby producing analgesic effects. Targeted drug delivery significantly enhances therapeutic efficacy across various diseases, particularly in cancer treatments, where conventional approaches such as chemotherapy and radiotherapy often cause severe side effects. In this context, nucleic acid aptamers-short, single-stranded DNA or RNA oligonucleotides capable of binding specific targets with high affinity-have emerged as promising tools for precision drug delivery and therapy. Aptamers can be selected against whole, living cells using SELEX and chemically modified for diverse applications.

Adolescence is a period of heightened neuroplasticity and vulnerability to environmental insults, including drug exposure. In this study, we investigated the short- and long-term behavioral effects, as well as the long-term hippocampal effects, of chronic cocaine administration during adolescence, along with the potential neuroprotective role of insulin-like growth factor 2 (IGF2) in male C57BL/6J mice. Over 21 days, mice received daily intraperitoneal injections of saline, cocaine, IGF2, or a combination of cocaine and IGF2.

Advances in understanding the pathophysiology of CRSwNP in relation to the predominant type 2 endotype have also paved the way for understanding possible overlaps with hypereosinophilic syndrome (HES) and eosinophilic granulomatosis with polyangiitis (EGPA). In this article, we present the biologic treatment options currently approved in Germany for the treatment of severe CRSwNP - dupilumab, omalizumab and mepolizumab - together with guidance on practical management including side effects for the indication of CRSwNP.
