Article Abstract

Increasing evidence supports the idea that intranuclear noncoding RNAs interact with target DNA to regulate their expression. The structural basis of this process involves the formation of DNA-RNA triple helices without unwinding the DNA double helix. This triplex formation process relies on the base pairing of the triplex target site (TTS) and the triplex-forming oligonucleotide (TFO), making the prediction of their binding crucial for mechanistic investigations. Here, we propose an online tool called Easy Triplex for predicting the formation of DNA-RNA triplexes. Using the canonical Hoogsteen pairing rules, Easy Triplex matches and predicts the details and distribution of all TTSs and TFOs on the basis of the user-provided DNA and RNA sequences. Practical application has shown that Easy Triplex can efficiently and accurately predict known triplex information. Its flexible parameter settings and scientifically set default output thresholds make Easy Triplex suitable for predicting the TFOs of both long and short noncoding RNAs. Additionally, users can choose to export the distribution information of CpG islands on the DNA sequence for further TTS screening. Thus, Easy Triplex is a simple and convenient online tool for predicting triplex formation. Easy Triplex is publicly accessible at http://easy-triplex.com/.

This work describes fully integrated multifolding electrochemical paper-based devices (ePADs) for enhanced multiplexed voltammetric determination of heavy metals (Zn(II), Cd(II), and Pb(II)) using tunable passive preconcentration. The paper devices integrate five circular sample preconcentration layers and a 3-electrode electrochemical cell. The hydrophobic barriers of the devices are drawn by pen-plotting with hydrophobic ink, while the electrodes are deposited by screen-printing.

Background: When diagnosing viral infections in humans and animals, the presence of virus in a sample in trace amounts that are below the analytical sensitivity of the detection system may cause false negative results and inaccurate diagnosis. We previously reported the development of a simple virion concentration technique using 12 ml large-volume samples that can dramatically improve diagnostic sensitivity by increasing analytical sensitivity by 100-fold over conventional methods. The present study was conducted to further improve the simplicity and versatility of this method.

A simple method has been developed for semi-quantitative analysis of the colorimetric output of loop-mediated isothermal amplification (LAMP) using a 3D-printed tube holder with a smartphone and notebook for the detection of Raillietina, which is the cause of Raillietiniasis affecting free-range chicken farming. In this method, a light is directed from a notebook screen to the LAMP products in the tube holder and the color absorption of the LAMP products is measured by using the appropriate smartphone application. It was found that the malachite green dye-coupled LAMP (MaG-LAMP) assay showed the highest sensitivity and specificity for detecting Raillietina without any cross-reaction with other related parasites and hosts.
