Smoking has been associated, among other factors, with musculoskeletal disorders. Although there is no consensus about the effects of smoking on osteoarthritis (OA), the increase in TNF-alpha in smokers has been considered an important factor in OA induction or progression. However, studies on the effects of smoking on chondrocytes are lacking. Here we aimed to study the effects of cigarette smoke extract (CSE) associated with a TNF-alpha inhibitor on cell death of primary human chondrocytes derived from osteoarthritic patients. CSE at 10% led to cell death by apoptosis after 48 h of incubation, together with caspase 3/7 activation, decrease in mitochondrial transmembrane potential, ROS production, and improvement in , , and gene expression. All these effects promoted by CSE were reversed by TNF-alpha inhibitor. Collagen II, F-actin, and SOX9 were also analyzed, and CSE promoted alteration in the expression of these proteins. In conclusion, our results support the clinical impact of smoking on OA development by showing the detrimental action of CSE on osteoarthritis-derived chondrocytes and the protective effects of TNF-alpha inhibitors, reinforcing the importance of this cytokine in the cartilage injury process.

Objectives: To investigate the effect of avitinib for suppressing NLRP3 inflammasome activation and alleviating septic shock and explore the underlying mechanism.
Methods: Mouse bone marrow-derived macrophages (BMDM), human monocytic leukemia cell line THP-1, and peripheral blood mononuclear cells (PBMC) isolated from healthy volunteers were pre-treated with avitinib, followed by activation of the canonical NLRP3 inflammasome using agonists including nigericin, monosodium urate (MSU) crystals, or adenosine triphosphate (ATP). Non-canonical NLRP3 inflammasome activation was induced intracellular transfection of lipopolysaccharide (LPS).

Ulcerative colitis (UC) is a persistent inflammatory condition marked by the destruction of the intestinal mucosal barrier, infiltration of inflammatory cells, and ulceration. M1/M2 macrophage polarization plays an imperative function in the regulation of inflammation through the nuclear factor-kappa B (NFκB) signaling pathway and modulating microRNA-155 (miR-155). Recent studies have highlighted the anti-ulcerogenic and colo-protective properties of sodium-glucose co-transporter-2 (SGLT2) inhibitors.

Lumbar spinal stenosis (LSS) is one of the most common spinal disorders in elderly people and is often accompanied by neuropathic pain. Although our previous studies have demonstrated that infiltrating macrophage contribute to chronic neuropathic pain in LSS rat model, the molecular mechanisms underlying macrophage activation and infiltration have not been fully elucidated. In this study, we examined the critical role of platelet-derived growth factor receptor (PDGFR) signaling pathway in neuropathic pain associated with macrophage infiltration and activation in LSS rats.

Post-traumatic stress disorder (PTSD) is a debilitating mental health condition stemming from exposure to traumatic events. Current treatment for PTSD is limited to the selective serotonin reuptake inhibitors, which are often associated with severe side effects and result in poor treatment adherence and limited effectiveness. Recent studies indicate that indoleamine 2,3-dioxygenase (IDO) may play a significant role in the development of stress-related disorders.

Neuroinflammation is vital in vincristine-induced peripheral neuropathy (VIPN). Locally infiltrated macrophages polarize to pro-inflammatory M1-type, release inflammatory cytokines, and contribute to neuropathic pain. Histone deacetylase 3 (HDAC3) regulates macrophage polarization.
