Nanoflow liquid chromatography-tandem mass spectrometry (nLC-MS) benefits untargeted metabolomics by enhancing sensitivity and integrating proteomics for the same sample. Here, we present a protocol to enable nLC-MS for dual metabolomics and proteomics. We describe steps for solid-phase micro-extraction (SPME)-assisted metabolite cleaning and enrichment, which avoids capillary column blockage. We then detail nLC-MS data acquisition and analysis. This protocol has been applied in diverse specimens including biofluids, cell lines, and tissues. For complete details on the use and execution of this protocol, please refer to Lin et al..

Given the widespread presence of imidacloprid in aquatic environments and the limited research on its impact on amphibian renal health, in this study, we investigated the effects of this commonly used neonicotinoid insecticide on kidney function and molecular mechanisms in Xenopus laevis. Employing a 28-day exposure model, histopathological changes and enzymatic responses induced by two concentrations of imidacloprid were examined, along with gene expression alterations and metabolic disruptions at environmentally relevant levels. The results highlighted significant renal histopathological damage and changes in key enzymes involved in oxidative stress and neurotoxicity, such as superoxide dismutase, glutathione S-transferase, and acetylcholinesterase.

Developing a practical strategy to enhance the quality of medicinal herb while alleviating negative plant-soil feedback (NPSF) is critical for agriculture. In this study, we investigated the effects of selenium nanoparticles (SeNPs) on Panax notoginseng through a two-year field experiment. Four treatments were established: a control (SeNPs_0) and three SeNPs concentrations (3, 5, and 10 mg/L), which were foliar-sprayed every 15 days for a total of six applications.

Dried blood spots (DBS) offer a practical and relatively non-invasive method for sample collection. Here, we evaluate the feasibility of applying H NMR spectroscopy to metabolomic analysis of DBS. Various solvent suppression techniques and extraction protocols were tested using aqueous and methanolic solvents.

Volvaria volvacea polysaccharides (VVP) possess diverse bioactivities with promising applications in biomedicine and functional foods. This study investigated the metabolic fate of VVP in human gut microbiota and uncovered the pivotal role of Bacteroides thetaiotaomicron using in vitro fecal fermentation models. VVP selectively promoted B.
