Herbivores are commonly classified as host specialists or generalists for various purposes, yet the definitions of these terms, and their intermediates, are often imprecise and ambiguous. We quantified host breadth for 240 non-native, tree-feeding insects in North America using phylogenetic diversity. We demonstrated that a partitioning of host breadth: (1) causes 67% of non-native insects to shift from a generalist to specialist category, (2) displays a reduction in host breadth from the native to introduced range, (3) identifies an inflection point in a model predicting the likelihood of non-native insect ecological impact, with a corresponding change in behaviour associated with specialists versus generalists, and (4) enables three models for strong prediction of whether a non-native forest insect will cause high impacts. Together, these results highlight the primacy of how herbivore host recognition and plant defences mediate whether novel host interactions will result in high impact after invasion.

Global efforts to standardise methodologies benefit greatly from open-source procedures that enable the generation of comparable data. Here, we present a modular, high-throughput nucleic acid extraction protocol standardised within the Earth Hologenome Initiative to generate both genomic and microbial metagenomic data from faecal samples of vertebrates. The procedure enables the purification of either RNA and DNA in separate fractions (DREX1) or as total nucleic acids (DREX2).

Most insect herbivores specialize on a few host plants; however, there are a minority of highly generalized species capable of feeding on hundreds of hosts. Generalism could emerge as a property of the species as a whole, while individuals would still exhibit greater specialization at more specific organizational levels. Yet, we lack studies with generalist insect herbivores directly testing this prediction.

Multiple bacterial immune systems, including CBASS, Thoeris, and Pycsar, employ signaling molecules that activate the immune response following phage infection. Phages counteract bacterial immune signaling using sponge proteins that bind and sequester the immune signals, but the breadth of immune signals targeted by phage sponges is unclear. Here we study the functional versatility of Acb2, Tad1 and Tad2, three families of sponge proteins known to inhibit CBASS and Thoeris signaling.

Dental calculus metagenomics has emerged as a valuable tool for studying the oral microbiomes of humans and a few select mammals. With increasing interest in wild animal microbiomes, it is important to understand how widely this material can be used across the mammalian tree of life, refine the related protocols and understand the expected outcomes and potential challenges of dental calculus sample processing. In this study, we significantly expand the breadth of studied host species, analysing laboratory and bioinformatics metadata of dental calculus samples from 32 ecologically and phylogenetically diverse mammals.

The 2015 Zika virus (ZIKV) outbreak in Brazil and its global spread underscored the urgent need for effective and broadly protective vaccines. While C57BL/6 and BALB/c mice are widely used in preclinical vaccine research, direct comparisons of their ability to elicit ZIKV-specific neutralizing antibodies (nAbs) remain limited. This study aimed to systematically evaluate and compare the immunogenic potential of these two common mouse strains across diverse vaccine platforms, focusing on their capacity to generate functional neutralizing antibody responses.
