Light simulations hold great potential for advancing optical techniques in neuroscience. They facilitate the in-silico refinement of optical stimulator designs and enable simulations of optical recordings from computational brain models, aiding neuroscience in forming a mechanistic understanding of brain circuitry. However, many published light models are inaccessible due to unavailable source code and documentation or are impractical due to excessive computational demands.

The Transient Receptor Potential Vanilloid sub-type 1 (TRPV1) is an ion channel that is activated by heat, extracellular protons, oxidation, and it is implicated in various aspects of inflammatory pain. In this study, we uncover that residue M308 in the TRPV1 ankyrin repeat domain (ARD) stands out from most other buried ARD residues because of the greater number of human missense variants at this position while maintaining a high degree of conservation across species and TRPV channel subtypes. We use mutagenesis and electrophysiology to examine this apparent discrepancy and show that substitutions at position M308 that preserve or reduce side-chain volume have no effect on channel function, whereas substitutions with larger or more polar residues increase channel activity in response to capsaicin or temperature.

Background: The International Standards for Neurological Classification of Spinal Cord Injury (ISNCSCI) is the most widely accepted system for characterizing sensorimotor impairments after spinal cord injury (SCI). There have been a number of ISNCSCI revisions, with the most recent edition published in 2019. Newer concepts, including the revised definitions of the zones of partial preservation (ZPPs) and documentation of non-SCI conditions, require training and practice for successful utilization.

Introduction: Projection radiography remains the most commonly used imaging modality in Irish hospitals. This study reviewed X-ray imaging protocols nationwide, assessing their structure, consistency, and alignment with clinical guidelines by comparing documentation, listed projections, and associated clinical indications.
Method: A descriptive comparative design was used to evaluate X-ray imaging protocols from 23 Model 3 and Model 4 Irish hospitals.
