Article Abstract

Global health is increasingly challenged by the growing prevalence of obesity and its associated complications. Quercetin, one of the most important dietary flavonoids, is being explored as an effective therapy for obesity with its mechanism remains understudied. Here in this study, it is demonstrated that quercetin intervention significantly reverses obesity-related phenotypes through reshaping the overall structure of microbiota, especially boosting colonization of the beneficial gut commensal Akkermansia muciniphila (A. muciniphila). Enrichment of A. muciniphila leads to generate more indole-3-lactic acid (ILA) to upregulate the expression of 12α-hydroxylase (CYP8B1) via fat mass and obesity-associated protein (FTO)/ N-methyladenosine (mA)/YTHDF2 manner, thereby facilitating cholesterol converts to cholic acid (CA). CA in turn drastically suppresses lipid accumulation via activating the farnesoid X receptor (FXR) in adipose tissue. This work introduces a novel therapeutic target for addressing obesity and expands upon the current limited understanding of the mediator function of mA modifications in microorganism-influenced bile acid (BA) metabolism.

Background & Aims: Among diet, microbiota, and obesity exists a close correlation that remains insufficiently explored, particularly within the pediatric age. We aimed to deeply investigate the relationship between dietary composition and microbiota in pediatric subjects with obesity before an educational training in a Mediterranean-style diet.

Methods: 55 subjects (10-18 years) with overweight or obesity and visceral adiposity, diet naïve, or failure to a previous weight loss program were phenotypically described through clinical and metabolic parameters, including circulating LPS levels.

Sunset yellow (SY) is a widely used food additive. However, its impacts on ulcerative colitis (UC) development remain unclear. Here, SY exposure exacerbated dextran sulfate sodium (DSS)-induced UC symptoms in mice, including body weight loss, elevated disease activity index, histological damage, inflammation, gut barrier impairment, disruption of gut microbiota composition, and sulfur metabolism.

Colorectal cancer (CRC) is a second leading cause of human mortality worldwide. Due to the inconsistent effectiveness of traditional anticancer therapies, there is an increasing need for novel drug-free cancer therapy that enhances the efficacy of current treatments with minimum side effects. CRC is linked to microbiome modifications, a phenomenon termed dysbiosis, which entails the reduction and/or augmentation of certain gut bacteria and their metabolites.

Probiotics have long been recognized for their health-promoting properties, primarily through their interaction with the gut microbiota. In recent years, Next-Generation Probiotics (NGPs), including and , have gained attention due to their potential therapeutic applications. Beyond live bacteria, the concept of postbiotics, defined as non-viable bacterial components with health benefits, has emerged, with membrane vesicles (MVs) representing a promising new class.
