This research focuses on developing and characterizing islatravir-loaded dissolving microarray patches (MAPs) to provide an effective, minimally invasive treatment option for human immunodeficiency virus (HIV-1) prevention and treatment. The research involves manufacturing these MAPs using a double-casting approach, and conducting in vitro and in vivo evaluations. Results show that the MAPs have excellent needle fidelity, structural integrity, and mechanical strength. in vitro studies demonstrate that the MAPs can penetrate skin up to 580 µm and dissolve within 2 hours. Permeation studies reveal that the delivery efficiency of islatravir across the skin is around 40%. In rodent models, these dissolving MAPs sustain islatravir delivery for up to 3 months. Scaling up the MAPs and increasing drug loading produced detectable levels in minipig. Projections from animal data suggest that these dissolving MAPs can achieve effective islatravir levels for a month after a single application in humans. These findings indicate dissolving MAPs as a minimally invasive approach to sustained release of islatravir.

Microneedle array patches (MAPs) offer a promising method for intradermal delivery, but conventional Co-Delivery MAPs (containing antigen and adjuvant together) have limited loading capacity and potential undesirable interactions. Adjuvants may also trigger adverse reactions in sensitive populations.

Traditional DNA vaccines, typically administered via intramuscular injection with electroporation (IM-E), often cause discomfort and require trained personnel. Addressing these challenges, we developed multivalent DNA vaccines targeting both intracellular mature virion (IMV) and extracellular enveloped virion (EEV) proteins of the monkeypox virus (MPXV), designated as M2 (A29L, B6R), M3 (A29L, B6R, M1R) and M4 (A29L, B6R, M1R, A35R). In this study, the water quality index (WQI) was calculated using multivariate statistics, incorporating physical, chemical, and microbiological analysis of water samples taken from water supply networks in the western district of Tehran from 2021 to 2024. The principal drinking water parameters such as pH, total hardness, turbidity, lead (Pb), chloride (Cl), fluoride (F), total dissolved solids (TDS), sulfate (SO), nitrate (NO), nitrite (NO), calcium (Ca), magnesium (Mg), arsenic (As), mercury (Hg), cadmium (Cd), fecal coliform and total residual chlorine (Ch) were selected according to Iranian national water standards. The WQI index was predicted using various machine learning algorithms, including multiple linear regression (MLR), support vector machine (SVM) regression, extreme gradient boosting (XGBoost), Random Forest (RF) regression, multilayer perceptron (MLP), and Kolmogorov-Arnold networks (KAN).
