Zika virus has raised global concerns due to its link to microcephaly and Guillain-Barré syndrome in adults. One of viral nonstructural proteins-NS4B, an integral membrane protein, plays crucial roles in viral replication by interacting with both viral and host proteins, rendering it an attractive drug target for antiviral development. We purified the N-terminal region of ZIKV NS4B (NS4B NTD) and reconstituted it into detergent micelles. Here, we report the assignments of the backbone resonances of NS4B NTD in detergent micelles. The available assignment is useful for understanding its structure and ligand binding to provide useful information for developing NS4B inhibitors.

Prokaryotes lack precise subcellular organelles but exhibit distinct regions such as cytoplasm, inner membrane, periplasm, and outer membrane, where most biochemical and physiological functions are organized. In this work, a simple method for the preparation of N-alkylated amino acid surfactants in 1-2 steps is reported. These products perform comparably to existing ionic surfactants, with N-tetrahydrogeranylated serine having a critical micelle concentration (CMC) of only 7.4 mmol·L.

The influence of poly(ethylene glycol) (PEG with molecular weights between 400 and 4000) on the critical micelle concentration (CMC) of nonionic detergents with maltose as well as oligo(ethylene glycol) head groups is determined by using 8-anilinonaphthalene-1-sulfonate (ANS) as fluorescence probe. The CMC is found to increase with the concentration of PEG (0%-30% (w/v)) in a way that is determined by the molar concentration of oxyethylene (OE) units and independent of the molecular weight of the added polymer. The effect is explained with the extended conformation of PEG in aqueous solution allowing for an interaction of detergent monomers with individual OE units via their alkyl tails.

Scramblases are membrane proteins that translocate phospholipids bidirectionally between the leaflets of a membrane bilayer. Examples of scramblases include Class A G protein-coupled receptors (GPCRs), members of the TMEM16 and DedA protein families, and protein insertases. To measure scramblase activity, the protein of interest is purified and reconstituted into large unilamellar vesicles that contain trace amounts of a fluorescent phospholipid reporter.
