Article Abstract

A class of side-chain type ferrocene macrocycles with a radially conjugated system is introduced in this study. The stereo configurations of these ferrocene rings were determined through single-crystal X-ray diffraction analysis. Notably, in the solid state, the ferrocene rings exhibit a distinctive herringbone stacking pattern imposed by a ferrocene-to-ring host-guest interaction. Through UV-vis absorption spectroscopy, electrochemical measurements, and theoretical calculations, valuable insights into the electronic properties of these rings were obtained. In addition, the single crystal of macrocycle demonstrates a second-order nonlinear optical response. As a class of organometallic nanorings, this work holds great potential for further exploration in the fields of organometallic chemistry, molecular electronics, and host-guest chemistry.

Previously undescribed steroids vernomigeodiins A-D (1-4), were isolated from the African medicinal plant Vernoniastrum migeodii along with known sterols 5-10 and the tripeptide aurantiamide acetate (11). The isolated steroids featured a stigmastane skeleton with a unique conjugated Δ-diene segment and a highly oxidized side chain, occasionally forming a bi- or tricyclic ring system. Sterols 1-3, 5-9 are glucosylated, whereas 4 and 10 are aglycons.

Nuclear factor erythroid 2-related factor 2 (Nrf2) and Kelch-like ECH-associated protein 1 (Keap1) axis is an attractive therapeutic target for various intractable diseases. Although protein-protein interaction inhibitors against Keap1-Nrf2 have been developed over the past decade, more structural expansion is needed to improve efficacy. In this study, linear polyethylenes bearing ketone groups in their side-chains were prepared by palladium-catalyzed copolymerization of ethylene and methyl vinyl ketone (poly(E/MVK)) and the photodegradation behavior was compared with polyethylene bearing in-chain carbonyl groups (poly(E/CO)). Poly(E/MVK) showed a faster photodegradation rate and a more significant decrease of the molecular weight than poly(E/CO). By H NMR analysis, both Norrish type I and type II scissions are involved for the degradation of poly(E/MVK), probably due to the existence of MVK groups in amorphous region, which is preferable for the radical chain reactions and the main chain cleavage.

The Transient Receptor Potential Vanilloid sub-type 1 (TRPV1) is an ion channel that is activated by heat, extracellular protons, oxidation, and it is implicated in various aspects of inflammatory pain. In this study, we uncover that residue M308 in the TRPV1 ankyrin repeat domain (ARD) stands out from most other buried ARD residues because of the greater number of human missense variants at this position while maintaining a high degree of conservation across species and TRPV channel subtypes. We use mutagenesis and electrophysiology to examine this apparent discrepancy and show that substitutions at position M308 that preserve or reduce side-chain volume have no effect on channel function, whereas substitutions with larger or more polar residues increase channel activity in response to capsaicin or temperature.
