Hydrogen peroxide (H2O2) transport by aquaporins (AQP) is a critical feature for cellular redox signaling. However, the H2O2 permeation mechanism through these channels remains poorly understood. Through functional assays, two Plasma membrane Intrinsic Protein (PIP) AQP from Medicago truncatula, MtPIP2;2 and MtPIP2;3 have been identified as pH-gated channels capable of facilitating the permeation of both water (H2O) and H2O2. Employing a combination of unbiased and enhanced sampling molecular dynamics simulations, we investigated the key barriers and translocation mechanisms governing H2O2 permeation through these AQP in both open and closed conformational states. Our findings reveal that both H2O and H2O2 encounter their primary permeation barrier within the selectivity filter (SF) region of MtPIP2;3. In addition to the SF barrier, a second energetic barrier at the NPA (asparagine-proline-alanine) region that is more restrictive for the passage of H2O2 than for H2O, was found. This behavior can be attributed to a dissimilar geometric arrangement and hydrogen bonding profile between both molecules in this area. Collectively, these findings suggest mechanistic heterogeneity in H2O and H2O2 permeation through PIPs.

Developing a promising on-demand controllable microneedle drug delivery system could provide stronger self-control and precision delivery of a large payload capacity. Nevertheless, the efficacy of existing systems has been constrained by limitations in the therapeutic payload capacity and slow diffusion of molecules, as well as the necessity for external resource configurations. Drawing inspiration from the multidimensional biomimetic strategies observed in the material properties and functional mechanisms of the bombardier beetle's defensive secretion system, a battery-free and self-propelled biomimetic microneedle system (BSBMs) is proposed for improving therapeutic outcomes and enabling controlled, on-demand drug delivery.

To investigate the inhibitory potential of plant-degraded polysaccharides on glycosidases and elucidate the mechanism by which they alleviate type 2 diabetes mellitus (T2DM), a degraded sweet corn cob polysaccharide (UHDSCP) was prepared using ultrasound-assisted hydrogen peroxide-ascorbic acid oxidation (HO/Vc). UHDSCP was subsequently characterized using various analytical techniques, including UV spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, gel permeation chromatography (GPC), high-performance liquid chromatography (HPLC), scanning electron microscopy (SEM), and atomic force microscopy (AFM). Microneedles have emerged as an effective strategy to bypass the stratum corneum by creating microchannels in the skin, allowing for enhanced drug permeation with minimal invasiveness. Pectin, a natural polysaccharide, when modified with thiol (-SH) groups, exhibits redox-sensitive behavior, making it responsive to reducing agents such as glutathione and dithiothreitol (DTT).
