Article Abstract

Rotaxanes are typically prepared using covalent bonds to trap a wheel component onto an axle molecule, and rotaxane-type wheel-axle assembly using only noncovalent interactions has been far less explored. Here we show that a dinickel(II) metallomacrocycle forms two different types of wheel-axle assemblies with a dibenzylammonium axle molecule based only on noncovalent interactions. The non-threaded assembly was obtained by introduction of Ni into the macrocycle before the complexation with the axle molecule (metal-first method). The non-threaded assembly was in rapid equilibrium with each of the components in solution. The threaded assembly was obtained by introduction of Ni after the formation of a pseudorotaxane from the non-metalated wheel and the axle molecule (axle-first method). The threaded assembly was not in equilibrium with the dissociated species even though it was maintained only by noncovalent interactions. Thus, formation of one of the non-threaded and threaded wheel-axle assemblies over the other is governed by the assembly pathway.

We report the deletion of nitrogen atoms from multiple template sites in rotaxanes, catenanes, and a molecular knot. Nitrogen extrusion from secondary amines in the backbone of the interlocked structures is achieved using -diphenylphosphinylhydroxylamine (DPPH), forming carbon-carbon bonds while largely maintaining the integrity of the original mechanical bonding. We find that DPPH gives improved yields (up to 51%) for nitrogen atom deletions from template sites in rotaxanes compared to an anomeric amide nitrogen-deletion reagent and overcomes a major substrate limitation in that, using DPPH, only one of the substituents of the secondary amine in the rotaxane axle needs to be radical-stabilizing.

Crown Ether-Peptide Rotaxanes.

We report on the metal-free active template synthesis of crown ether-peptide rotaxanes. A 24-crown-8 ring is sufficiently small that the side chains of canonical branched amino acids act as barriers that trap the macrocycle on the particular glycine residue used to assemble the rotaxane. The resulting crown ether-tripeptide rotaxane can subsequently be extended from either or both N- and C-termini of the axle.

The ability to control the relative motion between different components of molecules with precision is a cornerstone of synthetic nanotechnology. Mechanically interlocked molecules such as rotaxanes offer a platform for exploring this control by means of the positional manipulation of their components. Through imine condensation, a tetrahedral cage-shaped molecule was obtained in a one-pot manner, which comprises four trisamino vertices and four trisformyl faces each containing three -methyl-pyridyl units. Within the cage framework, each of the twelve pyridines undergoes pivoting around the corresponding imine-pyridine-phenyl axle, endowing the cage with adaptable conformations. By modulating the number of pyridyl nitrogen atoms that point inwards, the cage can switch the effective volume of its cavity where a wide range of guest molecules including quaternary ammoniums, or different amounts of alkali cations, are encapsulated, by utilizing dipole-cation forces and/or hydrogen bonding.
