Article Abstract

Despite neurobiological overlap, alcohol use disorder (AUD) and body mass index (BMI) show minimal genetic correlation (r), possibly due to mixed directions of shared variants. We applied MiXeR to investigate shared genetic architecture between AUD and BMI, conjunctional false discovery rate (conjFDR) to detect shared loci and their directional effect, Local Analysis of (co)Variant Association (LAVA) for local r, Functional Mapping and Annotation (FUMA) to identify lead single nucleotide polymorphisms (SNPs), Genotype-Tissue Expression (GTEx) to examine tissue enrichment, and BrainXcan to assess associations with brain phenotypes. MiXeR indicated 82.2% polygenic overlap, despite a r of -.03. ConjFDR identified 132 shared lead SNPs, with 53 novel, showing both concordant and discordant effects. GTEx analyses identified overexpression in multiple brain regions. Amygdala and caudate nucleus volumes were associated with AUD and BMI. Opposing variant effects explain the minimal rg between AUD and BMI, with implicated brain regions involved in executive function and reward, clarifying their polygenic overlap and neurobiological mechanisms.

Objective: Roux-en-Y gastric bypass (RYGB) has been linked with increased alcohol-related outcomes, whereas glucagon-like peptide-1 receptor agonists (GLP-1 RAs) have been associated with reduced risks. We performed the first multicenter retrospective cohort study to identify alcohol use outcomes of GLP-1 RAs post RYGB.

Given the potential for more rapid ethanol absorption in patients who have undergone bariatric surgery, we hypothesized that bariatric surgery is associated with risk for more severe alcohol-related liver disease. Biosensor-based, real-time stress detection has generated clinical interest for the purpose of driving just-in-time interventions that support recovery from mental disorders. Most stress detection models to date, however, have been trained with laboratory-based data from homogenous samples of healthy adults, and do not perform as well in clinical populations. As an initial step toward the development of a stress detection algorithm that functions well in clinical populations, we tested a series of stress-detection machine learning models on ambulatory electrocardiogram (ECG) and daily ecological momentary assessment (EMA) data collected from a sample of individuals in early recovery from alcohol use disorder (AUD).

Glucagon-like peptide 1 (GLP-1) receptor agonists, medications commonly employed in the treatment of type 2 diabetes mellitus, have illustrated several additional benefits, including weight loss and potentially reduce addictive cravings. Several studies have indicated that GLP-1 receptor agonists may be effective in treating Alcohol Use Disorder (AUD), for which current pharmacologic therapies are often inadequate. Proposed mechanisms include modulation of dopaminergic transmission and reduced gastric emptying, both of which reduce alcohol craving and tolerance.
