Article Abstract

Energetic stress compels cells to evolve adaptive mechanisms to adjust their metabolism. Inhibition of mTOR kinase complex 1 (mTORC1) is essential for cell survival during glucose starvation. How mTORC1 controls cell viability during glucose starvation is not well understood. Here we show that the mTORC1 effectors eukaryotic initiation factor 4E binding proteins 1/2 (4EBP1/2) confer protection to mammalian cells and budding yeast under glucose starvation. Mechanistically, 4EBP1/2 promote NADPH homeostasis by preventing NADPH-consuming fatty acid synthesis via translational repression of Acetyl-CoA Carboxylase 1 (ACC1), thereby mitigating oxidative stress. This has important relevance for cancer, as oncogene-transformed cells and glioma cells exploit the 4EBP1/2 regulation of ACC1 expression and redox balance to combat energetic stress, thereby supporting transformation and tumorigenicity in vitro and in vivo. Clinically, high EIF4EBP1 expression is associated with poor outcomes in several cancer types. Our data reveal that the mTORC1-4EBP1/2 axis provokes a metabolic switch essential for survival during glucose starvation which is exploited by transformed and tumor cells.

The small GTPase Rho5 has been shown to be involved in regulating the Baker's yeast response to stress on the cell wall, high medium osmolarity, and reactive oxygen species. These stress conditions trigger a rapid translocation of Rho5 and its dimeric GDP/GTP exchange factor (GEF) to the mitochondrial surface, which was also observed upon glucose starvation. We here show that rho5 deletions affect carbohydrate metabolism both at the transcriptomic and the proteomic level, in addition to cell wall and mitochondrial composition.

Purpose: This study aimed to develop a composite nanozyme system (Au/PB-Ce6-HA) based on gold nanoparticles (AuNPs) and Prussian blue nanoparticles (PBNPs) to combat tumor hypoxia and insufficient endogenous hydrogen peroxide (HO) deficiency, thus enhancing the efficacy of sonodynamic therapy (SDT) and starvation therapy for liver cancer.

Methods: The Au/PB-Ce6-HA system was constructed by in situ embedding AuNPs on PBNPs, loading the sonosensitizer Chlorin e6 (Ce6), and surface-coating with thiolated hyaluronic acid (HA-SH). The system was evaluated both in vitro and in vivo to assess its ability to catalyze glucose to generate HO, decompose HO to produce oxygen, and generate highly toxic reactive oxygen species (ROS) under ultrasound irradiation.

Abnormal glycolysis and glutamine metabolism not only sustain tumor growth but also reprogram the tumor microenvironment (TME). However, due to compensatory mechanisms and low tumor immunogenicity, targeting a single metabolic pathway is often insufficient for effective cancer therapy. Tumour progression depends on the bidirectional interactions between cancer and stroma in the heterogeneous tumour microenvironment (TME) partially through extracellular vesicles (EVs). However, the secretary mechanism and biological effect of cancer cell derived EVs on tumour survival under starvation is poorly defined. Here, we identify cancer cells selectively secrete miR-33a with the assistance of aconitase 1 (ACO1), an iron-responsive RNA binding protein, under glucose starvation and lower iron level, which affiliates the binding capability of miR-33a and ACO1.

Proteasomes are large multiprotein complexes required for selective intracellular protein degradation, regulating numerous cellular processes and maintaining protein homeostasis and organismal health. In the budding yeast Saccharomyces cerevisiae grown under different glucose conditions, proteasomes undergo dynamic phase transitions between free and condensate states concomitant with nucleocytoplasmic translocation.
