Article Abstract

Neurons on the left and right sides of the nervous system often show asymmetric properties, but how such differences arise is poorly understood. Genetic screening in zebrafish revealed that loss of function of the transmembrane protein Cachd1 resulted in right-sided habenula neurons adopting left-sided identity. Cachd1 is expressed in neuronal progenitors, functions downstream of asymmetric environmental signals, and influences timing of the normally asymmetric patterns of neurogenesis. Biochemical and structural analyses demonstrated that Cachd1 can bind simultaneously to Lrp6 and Frizzled family Wnt co-receptors. Consistent with this, mutant zebrafish lose asymmetry in the habenulae, and epistasis experiments support a role for Cachd1 in modulating Wnt pathway activity in the brain. These studies identify Cachd1 as a conserved Wnt receptor-interacting protein that regulates lateralized neuronal identity in the zebrafish brain.

Investigating the impact of hyperbilirubinemia on cognitive dysfunction in adult zebrafish: an in vivo model.

Korean J Anesthesiol

Background: Despite the well-known effects of elevated bilirubin in neonates, its neurotoxic potential in adults remains uncertain. In perioperative and hepatic disease contexts, transient bilirubin elevations are common; however, their direct contribution to cognitive dysfunction has not been clearly established. This study aimed to determine whether transient bilirubin elevation alone can impair cognition and disrupt blood-brain barrier (BBB) function in adult zebrafish, and to compare these effects with those of liver injury.

The SH2B family, which includes SH2B1, SH2B2, and SH2B3, consists of adaptor proteins that possess conserved Src homology 2 (SH2) and pleckstrin homology (PH) domains, playing essential roles as signaling mediators. However, the gene expression patterns of this family during embryonic development are still mostly unclear. In this study, we first investigated the evolutionary conservation of SH2B across multiple species using phylogenetic analysis, which revealed high sequence homology between zebrafish Sh2b and its orthologs in other vertebrates.

Synaptic plasticity is fundamental for cognitive development and brain function. Aluminium nanoparticles (AlNPs), widely used in industrial and consumer products, pose potential neurotoxic risks, particularly during early neurodevelopment. However, their effects on synaptic plasticity and cognitive outcomes remain poorly understood.

Developmental epileptic encephalopathies (DEEs), including Dravet syndrome (DS), require antiseizure medications (ASMs) that balance efficacy with developmental safety. There is an urgent clinical need for novel therapeutic agents that combine potent anticonvulsant activity with developmental safety. β-Asarone, an active constituent of plants, has demonstrated antiepileptic potential, but its toxicities severely limit clinical application.

Early diagnosis is critical for the effective management of neurodegenerative disorders, and retinal alterations have emerged as promising early biomarkers due to the retina's close developmental and functional link to the brain. The zebrafish (Danio rerio), with its rapid development, transparent embryos, and evolutionarily conserved visual system, represents a powerful and versatile model for studying retinal degeneration. This review discusses a range of behavioral assays-including visual adaptation, motion detection, and color discrimination-that are employed to evaluate retinal function in zebrafish.
