Transient receptor potential vanilloid family member 1 (TRPV1) has been revealed as a therapeutic target of osteoarthritis (OA), the most common deteriorating whole joint disease, by impeding macrophagic inflammation and chondrocytes ferroptosis. However, the clinical application for capsaicin as the TRPV1 agonist is largely limited by its chronic toxicity. To address this issue, we developed a bifunctional controllable magnetothermal switch targeting TRPV1 for the alleviation of OA progression by coupling of magnetic nanoparticles (MNPs) to TRPV1 monoclonal antibodies (MNPs-TRPV1). Under the alternating magnetic field (AMF) stimulation, MNPs-TRPV1 locally dissipated heat, which was sufficient to trigger the opening and activation of TRPV1, and effectively impeded macrophagic inflammation and chondrocyte ferroptosis. This magnetothermal modulation of TRPV1 simultaneously attenuated synovitis and cartilage degeneration in mice incurred by destabilization of medial meniscus surgery, indicating the delayed OA progression. Furthermore, MNPs-TRPV1 with AMF exposure remarkably reduced knee pain sensitivity, alleviated the crippled gait, and improved spontaneous ambulatory activity performance in the mice OA model. Overall, this work provides a potential pathogenesis-based precise OA therapy with temporally and spatially magnetothermal modulation of TRPV1 in a controllable manner.

In this study, we analyze the thermal processes in Ising-type exchange-coupled binuclear clusters induced by the sudden switching off of an initially applied magnetic field. The main result is the established correlation between the sign of the exchange parameter and the sign of the thermal effect caused by the fast field switching off. We demonstrate that in the case of ferromagnetic Ising-type superexchange associated with easy-axis magnetic anisotropy, the spin system absorbs heat, resulting in the cooling of the surrounding.

Spatiotemporal regulation of therapeutic enzymes is desirable for enhancing the efficacy and safety of enzyme-based treatments for metabolic diseases, yet the absence of techniques capable of on-demand manipulating the in vivo catalytic activity of urate oxidase (UOx) represents a significant challenge in achieving precise gout therapy. Herein, we report a cyclic cascade nanohybrid urate oxidase (NUOx) comprised of a FeO nanoring core and a UOx shell, whose activity can be switched on and off on-demand using a deep-penetrated alternating magnetic field (AMF). The FeO nanoring under AMF exposure functions as a nanoheater to stimulate its intrinsic catalase (CAT) activity for oxygen recycling, which in turn activates UOx/CAT cascade for controlled uric acid degradation.

Tumor recurrence and wound infection are devastating complications of wide excision surgery for melanoma, and deep postoperative wound drainage typically increases pain. An amoeba-inspired magnetic soft robot (ASR) with switchable dormant and active phases is developed to address the aforementioned challenges. The dormant ASR supports wounds through its solid-like elasticity and regulates reactive oxygen species (ROS) levels bidirectionally, promoting healing in infected wounds and eliminating residual tumors.
