Article Abstract

General and efficient strategies for highly diastereoselective synthesis of divergent heterocyclic scaffolds through desymmetric [3+3] cycloaddition of -quinamines with 1,3-dipole surrogates hydroximoyl halides and α-halohydroxamates have been developed. This synthetic protocol provided a variety of heterocyclic architectures containing 1,2,4-oxadiazine and hydroquinoxaline skeletons in good yields with a wide substrate scope.

The first enantioselective Bora-Brook rearrangement reaction catalyzed by a Cu/SOP system was established. In this protocol, the synthesis of axially chiral monoaldehydes has been achieved by the desymmetric reduction of prochiral dialdehydes. This reaction proceeds smoothly under mild conditions, affording the products in high yields (up to 91%) with excellent enantioselectivities (up to 96:4 er) while exhibiting broad functional group compatibility.

Di(hetero)aroyl dichlorides are desymmetrized upon sequential reaction with alcohols and 2-methyl N-benzyl thiazolium salts within the course of a one-pot three-component reaction yielding ester-substituted aroyl-S,N-ketene acetals under mild conditions in good yields. A prerequisite for the concise one-pot process is the different nucleophilicity of the alcohols and in situ generated S,N-ketene acetals. The resulting compounds are merocyanines with dominant charge-transfer absorption bands which are fluorescent in the solid state, but not in solution.

The generation of metal carbenes from readily available allenes represents a remarkable advance in metal carbene chemistry. However, most of these transformations are mainly restricted to the noble-metal catalysts (Au and Pt). Here, a copper-catalyzed desymmetric cyclization reaction of divinylallenes is described, enabling the practical and atom-economical synthesis of a diverse array of valuable triazolo-fused pyridazines and tetracyclic N-heterocycles by a presumable copper carbene intermediate and a highly selective 1,2-N shift process.

The incorporation of enantioenriched difluoromethyl cyclopropane (DFC) groups into drug candidates has garnered increasing attention in the pharmaceutical industry due to the unique ability of the DFC groups to serve as conformationally rigid hydrogen-bond donors. Despite their potential, the widespread use of chiral DFC groups has been limited by their challenging synthesis. Here we report the use of difluoromethyl-copper complexes for the development of a desymmetric difluoromethylation reaction, using enantioselective copper catalysis.

In this study, a series of unsymmetrical single-benzene fluorophore (SBF) derivatives were systematically synthesized by selectively disrupting the ester symmetry of standard diamino terephthalate-type analogs alternative synthetic routes. Various electron-donating groups (EDGs) and electron-withdrawing groups (EWGs) were introduced at one ester group position, yielding 12 A series compounds with emission wavelengths spanning from 440 to 578 nm. EDGs led to blue-shifts, while EWGs induced red-shifts relative to the symmetric parent compound.
