Curcumin is a natural phenylpropanoid compound with various biological activities and is widely used in food and pharmaceuticals. A curcumin biosynthetic pathway was constructed in BL21(DE3). Optimization of the curcumin biosynthesis module achieved a curcumin titer of 26.8 ± 0.6 mg/L. Regulating the metabolic fluxes of the β-oxidation pathway and fatty acid elongation cycle and blocking the endogenous malonyl-CoA consumption pathway increased the titer to 113.6 ± 7.1 mg/L. Knockout of endogenous curcumin reductase () and intermediate product detoxification by heterologous expression of the solvent-resistant pump () increased the titer to 137.5 ± 3.0 mg/L. A 5 L pilot-scale fermentation, using a three-stage pH alternation strategy, increased the titer to 696.2 ± 20.9 mg/L, 178.5-fold higher than the highest curcumin titer from biosynthesis previously reported, thereby laying the foundation for efficient biosynthesis of curcumin and its derivatives.

Importance: Merkel cell carcinoma (MCC) is typically caused by the Merkel cell polyomavirus (MCPyV) and recurs in 40% of patients. Half of patients with MCC produce antibodies to MCPyV oncoproteins, the titers of which rise with disease recurrence and fall after successful treatment.
Objective: To assess the utility of MCPyV oncoprotein antibodies for early detection of first recurrence of MCC in a real-world clinical setting.

Objective: To evaluate dynamic changes in autoantibody and proteomic profiles in treatment-naïve systemic sclerosis (SSc) patients and identify biomarkers and mechanisms associated with disease progression.
Methods: Serum samples from 30 baseline and 49 follow-up SSc patients, along with 38 controls, were analyzed. Autoantibody profiles were assessed using an autoantigen microarray targeting 120 autoantibodies, while proteomic analysis was conducted via liquid chromatography-mass spectrometry in data-independent acquisition mode.

This report presents the follow-up of a patient in whom we assumed mild NMDA-R encephalitis in the first psychotic episode.

Objective: Our objective was to estimate the seroprevalences of 6 serovars in beef calves at or near fall weaning and assess how concentrations of serovar antibody titers in weaning-age calves varied with herd vaccination programs.
Animals: Serum was collected from 1922 beef calves from 106 herds in the Canadian Cow-Calf Surveillance Network (C3SN).
Procedure: A microscopic agglutination test was used to measure antibody titers for serovars Bratislava, Canicola, Grippotyphosa, Hardjo, Icterohaemorrhagiae, and Pomona.
