Fisetin is a flavonoid found in plants and has been reported to be effective in various human diseases. However, the effective mechanisms of ultraviolet-A (UVA)-mediated skin damage are not yet clear. In this study, we investigated the protective mechanisms of fisetin regarding UVA-induced human dermal fibroblasts (HDFs) and human epidermal keratinocytes (HEKs) damages. Fisetin showed a cytoprotective effect against UVA irradiation and suppressed matrix metalloproteinases (MMPs), MMP-1, and MMP-3 expression. In addition, fisetin was rescued, which decreased mRNA levels of pro-inflammatory cytokines, reactive oxygen species production, and the downregulation of MAPK/AP-1 related protein and NADPH oxidase (NOX) mRNA levels. Furthermore, UVA-induced MMP-1 and MMP-3 were effectively inhibited by siRNAs to NOX 1 to 5 in HDFs and HEKs. These results indicate that fisetin suppresses UVA-induced damage through the NOX/ROS/MAPK pathway in HDFs and HEKs.

Selective inhibitors are essential for targeted therapeutics and for probing enzyme functions in various biological systems. The two main challenges in identifying such protein-based inhibitors lie in the extensive experimental effort required, including the generation of large libraries, and in tailoring the selectivity of inhibitors to enzymes with homologous structures. To address these challenges, machine learning (ML) is being used to improve protein design by training on targeted libraries and identifying key interface mutations that enhance affinity and specificity.

Purpose: This study aimed to investigate the causal relationship between matrix metalloproteinases (MMPs) and cervical lesions, as their correlation remains unclear despite reports of their involvement in the pathogenesis of these lesions.
Patients And Methods: A two-sample Mendelian randomization (MR) analysis was performed using genome-wide association studies (GWAS) summary data to assess the causal associations of MMPs (MMP-1, 2, 3, 7, 9, 10, 12, 14, 16, and 17) with cervical lesions. The primary analysis utilized the inverse-variance weighted (IVW) method, supplemented by weighted median, mode, and MR-Egger regression.

Objective: Aim: To study peculiarities of the state of extracellular matrix in testicular embryonal carcinoma.
Patients And Methods: Materials and Methods: Samples of testicular embryonal carcinoma tissue were collected from 3 groups of patients (group «1» - stages T1N0S0-2, group «2» - T2N1-3S0-2, group «3» - T2-3N0-3S0-2 with distant metastases) to assess the state of its extracellular matrix investigating MMP-1, MMP-3 and MMP-9 expression.
Results: Results: The levels of MMP-1, MMP-3 and MMP-9 expression were statistically significantly higher in patients with tumorous vascular/lymphatic invasion, metastases in regional lymph nodes, distant metastases and invasion into the spermatic cord (except of lightness of MMP-3 expression) in comparison with patients without these characteristics.

Background: This study reveals elevated levels of MMP-1, MMP-2, and MMP-3 in endometriotic lesions, potentially reflecting a senescenceassociated secretory phenotype. No corresponding increase was observed in peritoneal fluid, possibly due to technical or sample-related limitations. These findings support the potential of MMPs as biomarkers and therapeutic targets in endometriosis.
