Article Abstract

Elevated impulsivity is a key component of attention-deficit hyperactivity disorder (ADHD), bipolar disorder and juvenile myoclonic epilepsy (JME). We performed a genome-wide association, colocalization, polygenic risk score, and pathway analysis of impulsivity in JME (n = 381). Results were followed up with functional characterisation using a drosophila model. We identified genome-wide associated SNPs at 8q13.3 (P = 7.5 × 10) and 10p11.21 (P = 3.6 × 10). The 8q13.3 locus colocalizes with SLCO5A1 expression quantitative trait loci in cerebral cortex (P = 9.5 × 10). SLCO5A1 codes for an organic anion transporter and upregulates synapse assembly/organisation genes. Pathway analysis demonstrates 12.7-fold enrichment for presynaptic membrane assembly genes (P = 0.0005) and 14.3-fold enrichment for presynaptic organisation genes (P = 0.0005) including NLGN1 and PTPRD. RNAi knockdown of Oatp30B, the Drosophila polypeptide with the highest homology to SLCO5A1, causes over-reactive startling behaviour (P = 8.7 × 10) and increased seizure-like events (P = 6.8 × 10). Polygenic risk score for ADHD genetically correlates with impulsivity scores in JME (P = 1.60 × 10). SLCO5A1 loss-of-function represents an impulsivity and seizure mechanism. Synaptic assembly genes may inform the aetiology of impulsivity in health and disease.

The sterile insect technique (SIT) is a highly effective biologically-based method for the suppression of many insect pest populations. SIT efficacy could be improved by methods of male sterilization that avoid the use of irradiation that can result in diminished fitness and mating competitiveness. Alternative sterilization methods include conditional disruption of genes for male fertility, or using their sperm-specific promoters to drive the expression of genes for lethal effectors.

Sauvagesia rhodoleuca is an endangered species endemic to southern China. Due to human activities, only six fragmented populations remain in Guangdong and Guangxi. Despite considerable conservation efforts, its demographic history and evolution remain poorly understood, particularly from a genomic perspective.

Ring finger protein 180 (RNF180) is an E3 ubiquitin-protein ligase that promotes polyubiquitination and degradation. We analyzed the roles and molecular mechanisms of RNF180 during the tumorigenesis and progression of colorectal cancer (CRC) through bioinformatics analysis, in vivo and vitro experiments. RNF180 overexpression was observed in CRC, and positively associated with T, N and TNM staging or differentiation.

This study investigated the effects of five representative biocarriers-biochar (BC), activated carbon (AC), nano-magnetite (NM), zero-valent iron (ZVI), and polyurethane sponge (PUS)-on chain elongation (CE) from ethanol/acetate in anaerobic systems. All carriers enhanced CE to varying extents. BC and NM significantly increased caproate yields (6032.

Bardet-Biedl Syndrome (BBS) is a rare autosomal recessive ciliopathy characterized by genetic heterogeneity. Despite significant progress in understanding the BBSome-coding genes associated with ciliopathies, the pathogenesis linked to mutations in chaperonin-coding genes (BBS6, BBS10, and BBS12) remains poorly defined. This study aims to confirm the genetic diagnosis of BBS and elucidate the pathological mechanisms in causative genes of BBS10 and BBS12.
