Cytoplasmic poly(A)-binding protein (PABPC; Pab1 in yeast) is thought to be involved in multiple steps of post-transcriptional control, including translation initiation, translation termination, and mRNA decay. To understand these roles of PABPC in more detail for endogenous mRNAs, and to distinguish its direct effects from indirect effects, we have employed RNA-Seq and Ribo-Seq to analyze changes in the abundance and translation of the yeast transcriptome, as well as mass spectrometry to assess the abundance of the components of the yeast proteome, in cells lacking the gene. We observed drastic changes in the transcriptome and proteome, as well as defects in translation initiation and termination, in Δ cells. Defects in translation initiation and the stabilization of specific classes of mRNAs in Δ cells appear to be partly indirect consequences of reduced levels of specific initiation factors, decapping activators, and components of the deadenylation complex in addition to the general loss of Pab1's direct role in these processes. Cells devoid of Pab1 also manifested a nonsense codon readthrough phenotype indicative of a defect in translation termination, but this defect may be a direct effect of the loss of Pab1 as it could not be attributed to significant reductions in the levels of release factors.

Study Objectives: This study compares the demographic characteristics and experiences of three different groups of adolescent and young adults (AYAs) seeking non-sedated intrauterine device (IUD) insertion in the outpatient setting: (1) cisgender individuals, (2) transgender and gender diverse (TGD) individuals receiving gender-affirming testosterone therapy, and (3) TGD individuals not receiving gender-affirming testosterone therapy.
Methods: This retrospective cohort study included AYAs age 13 to 21 years seeking IUD insertion. Data were obtained from the electronic health record via a pre-existing, multi-institutional shared quality improvement (QI) collaborative.

Background: Select patients with metastatic clear-cell renal-cell carcinoma can be treated without systemic therapy, yet few studies have explored this population. We investigated the efficacy of metastasis-directed therapy without systemic therapy in oligometastatic clear-cell renal-cell carincoma.
Methods: This investigator-initiated single-arm, phase 2 trial enrolled patients aged 18 years or older with an Eastern Cooperative Oncology Group performance status of 0-2, histologically confirmed clear-cell renal-cell carcinoma, and one to five metastases.

To optimize the deployment of Generative Artificial Intelligence in health care, it's essential for health care professionals (HCPs) to understand these technologies' capabilities and constraints. This study explores HCPs' initial impressions and experiences using ChatGPT, a Generative Pre-trained Transformer, in Pediatric Critical Care Units (PICUs). By conducting focus groups with a diverse set of HCPs, we aimed to assess their awareness, utilization, perceived benefits, and concerns about incorporating ChatGPT into their PICUs.
