Methods for sensitive and high-throughput evaluation of CRISPR RNA-guided nucleases (RGNs) off-targets (OTs) are essential for advancing RGN-based gene therapies. Here we report SURRO-seq for simultaneously evaluating thousands of therapeutic RGN OTs in cells. SURRO-seq captures RGN-induced indels in cells by pooled lentiviral OTs libraries and deep sequencing, an approach comparable and complementary to OTs detection by T7 endonuclease 1, GUIDE-seq, and CIRCLE-seq. Application of SURRO-seq to 8150 OTs from 110 therapeutic RGNs identifies significantly detectable indels in 783 OTs, of which 37 OTs are found in cancer genes and 23 OTs are further validated in five human cell lines by targeted amplicon sequencing. Finally, SURRO-seq reveals that thermodynamically stable wobble base pair (rG•dT) and free binding energy strongly affect RGN specificity. Our study emphasizes the necessity of thoroughly evaluating therapeutic RGN OTs to minimize inevitable off-target effects.

To investigate the anti-relaxation performance of FOTS-modified OTS coatings on the inner walls of cesium (Cs) atomic cell, this study employs molecular dynamics (MD) simulations to explore the self-assembly process of FOTS-modified OTS molecular chains on the SiO (001) surface and evaluates the effects of FOTS chain amounts, water molecule content, and temperature on the diffusion behavior of Cs atoms. Results show that the optimized interface model of the FOTS-modified OTS coating and SiO substrate achieves thermodynamic and energetic equilibrium under the conditions of 25 °C and 2000 ps. The film formation process of FOTS-modified OTS chains on SiO surfaces involves three distinct stages: initial anchoring, conformational rearrangement, and structural relaxation and equilibrium configuration.

Reactions of a cerium(III) o-,-dimethylaminobenzyl (DMAB) complex with oxidizing agents, dichalcogenides, and a thiophosphinic amide have been studied. Treatment of the reported homoleptic DMAB complex [Ce(DMAB)] () with Ag(OTs) (OTs = tosylate) and Ag(OTf) (OTf = triflate) in tetrahydrofuran (thf) afforded [Ce(DMAB)(μ-OTs)(thf)] () and [Ce(DMAB)(OTf)(thf)] (), respectively. Reactions of with CuI and CuCl led to isolation of [Ce(DMAB)I] () and [Cu(DMAB)], respectively.

While Te-based ovonic threshold switching (OTS) materials offer advantages such as low-voltage operation and fast switching speed, their relatively low crystallization temperature compared to S- or Se-based counterparts results in poor thermal stability and limited electrical endurance. Various strategies, including element doping and complex composition design, have been explored to address these limitations. In this study, the OTS device properties of BTe thin films, a simple two-component system, were systematically investigated across a wide range of compositions.
