Article Abstract

Background/aim: Clear-cell renal cell carcinoma (ccRCC) is a common recalcitrant cancer. However, little is known about biomarkers of ccRCC. In the present study, we investigated the role of guanylate-binding protein 2 (GBP2), an interferon-induced GTPase, in ccRCC and its potential as a biomarker of this disease.

Materials And Methods: GBP2 expression was analyzed using the Gene Expression Omnibus, The Cancer Genome Atlas, and Human Protein Atlas databases. Univariate and multivariate Cox-regression analyses were used to investigate the prognostic value of GBP2 in ccRCC. In addition, quantitative real-time polymerase chain reactions, western blotting, and immunohistochemistry were performed to confirm the expression of GBP2 in tissues from patients with ccRCC who had undergone radical nephrectomy at the Department of Urology, the First Affiliated Hospital of Chongqing Medical University (Chongqing, P. R. China). Cell-function assays were performed to investigate the effect of GBP2 on Caki-1 human kidney-cancer cells.

Results: Bioinformatics and in vitro experiments showed that the expression of GBP2 was up-regulated in ccRCC tissues and cells and was positively correlated with the malignant clinicopathological parameters of the disease. Univariate and multivariate Cox-regression analyses showed that GBP2 expression was an independent risk factor for the prognosis of ccRCC. Cell-function assays showed that GBP2 expression promoted the proliferation, migration, and invasion of Caki-1 cells through the WNT/β-catenin signaling pathway.

Conclusion: The present results indicate that GBP2 expression may serve as a prognostic biomarker for ccRCC.

Mutations in the histone methyltransferase enhancer of zeste homolog 2 (EZH2), particularly the neomorphic Y641F hot spot mutation, are implicated in hematologic malignancies. However, how developmental timing and cellular context influence their oncogenic potential remains poorly understood. Here, we used a conditional allele with multiple tissue-specific Cre drivers to investigate the effects of these mutations across hematopoietic development.

The interferon (IFN)-inducible GTPases play a crucial role in cell autonomous immunity against intracellular pathogens. Particularly, these GTPases directly recognize the host membrane-derived vacuole containing pathogens and subsequently destroy it. Although it has been revealed that these GTPases target the membrane of ()-containing vacuole (LCV), molecular mechanism has been totally uncleared.

Neobaicalein, a natural flavonoid compound, has shown potential therapeutic effects on various neurodegenerative diseases. However, its role and the underlying mechanisms in retinal damage remain largely unexplored. A mouse model of acute ocular hypertension (AOH) was established by raising intraocular pressure to 90 mmHg for 60 min.

Introduction: Fine particulate matter (PM) is an important factor in the induction of a variety of respiratory diseases and associated cellular damage. The composition of PM in the animal farm environments is complex, which poses a significant threat to the respiratory health of both workers and livestock, but the causative mechanisms are unclear.

Methods: In order to investigate targeted treatment options, this study focused on the role of microbial components in cowshed PM-induced respiratory damage.

Objective This study aims to investigate the expression, phenotypic changes, and mechanisms of action of guanylate-binding protein 2 (GBP2) in the process of silica-induced pulmonary fibrosis. Methods The expression and localization of GBP2 in silicotic lung tissue were detected by immunohistochemical staining and immunofluorescence. An in vitro cell model was constructed, and methods such as Western blot and real-time quantitative reverse transcription polymerasechain reaction were utilized to investigate the function of GBP2 in different cell lines following silica stimulation.
