Background: The emergence of drug-resistant pathogens has stimulated the need for the development of new antimicrobial agents. Epigenetic modulation by suppressing epigenetic inhibitors, such as 5-azacytidine (5-aza), has been shown to activate silent biosynthetic gene clusters within a fungus and causes the production of novel secondary metabolites. This research examined this epigenetic modification strategy in the poorly studied filamentous fungus, Ceratorhiza hydrophila, which may help induce the additional production of bioactive compounds.

Profile of NT-0527, a brain penetrant NLRP3 Inflammasome inhibitor suitable as an tool compound for neuroinflammatory disorders.

RSC Med Chem

September 2025

NodThera Ltd. Suite 8, The Mansion, Chesterford Research Park, Little Chesterford, Saffron Walden Essex CB10 1XL UK

Inhibition of the NLRP3 inflammasome has emerged as a high potential treatment paradigm for the treatment of neuroinflammation, with demonstrated anti-neuroinflammatory effects in Parkinson's disease patients and a strong rationale in Alzheimer's disease and amyotrophic lateral sclerosis. To facilitate further progress in this field, brain penetrant NLRP3 inflammasome inhibitors as leads and tool compounds are required. We discovered a small molecule NLRP3 inflammasome inhibitor, NT-0527 (11), and extensively profiled this to reveal a highly potent, selective and brain penetrant compound.

Amyloid-β (Aβ) is implicated in the pathophysiology of Alzheimer's disease (AD) and plays a significant role in neuronal degeneration. Aβ in solution is essential during the initial stages of developing lead compounds that influence Aβ fibrillation. The tendency of the Aβ peptide to misfold in solution is correlated with the etiology of AD.

Objective: To evaluate the clinical efficacy of tocilizumab, a interleukin-6 (IL-6) receptor blocker, for the treatment of acute necrotizing encephalopathy (ANE).

Methods: PubMed, Cochrane Library, Embase, and Web of Science were searched for systematic review based on PRISMA guidelines. ANE patients treated with and without tocilizumab were included.

Voxel-level Radiomics and Deep Learning Based on MRI for Predicting Microsatellite Instability in Endometrial Carcinoma: A Two-center Study.

Acad Radiol

September 2025

Department of Radiology, Air Force Medical Center, Air Force Medical University, Fucheng Road 30, Haidian District, Beijing 100142, China (P.S., S.L., N.X.). Electronic address:

Rationale And Objectives: To develop and validate a non-invasive deep learning model that integrates voxel-level radiomics with multi-sequence MRI to predict microsatellite instability (MSI) status in patients with endometrial carcinoma (EC).

Methods: This two-center retrospective study included 375 patients with pathologically confirmed EC from two medical centers. Patients underwent preoperative multiparametric MRI (T2WI, DWI, CE-T1WI), and MSI status was determined by immunohistochemistry.
