Article Abstract

"San Huang Powder," a nonsterile milled herb powder, is frequently used to treat burn wounds in traditional Chinese herbal medicine. However, treating a wound with a nonsterile dressing or reagent is not compatible with the current guidelines in modern medicine. Therefore, we investigated the bactericidal and anti-inflammatory activities of four herb extracts used in "San Huang Powder" . Meanwhile, an porcine model with superficial second-degree burns was used for the experiments since the size and skin composition of pigs are the closest to that of the human body. The minimal bactericidal concentration (MBC) of the herb extracts was determined. The assay indicated that Rhubarb and Phellodendron bark extracts decreased the levels of inflammatory cytokines, IL-8, and GM-CSF on LPS-induced HMEC-1 cells. In accordance with this result, the histopathological evaluation results showed that the efficacy of "San Huang Powder" containing both herb materials was much better than the group without Rhubarb. Our results not only provide a basis to understand why "San Huang Powder" has been used to clinically treat wounds without sterilization directly since ancient times but also show the advantages of using multiple herb materials simultaneously on wound sites to prevent infection during treatment. Rhubarb is the recommended ingredient involved in the preparation of "San Huang Powder" to ensure the healing efficacy of burn wounds.

Metastasis, particularly in aggressive triple-negative breast cancer (TNBC), is the leading cause of breast cancer-related mortality. San-Huang-Xie-Xin-Tang (SHXXT), a heat-clearing herbal formula, is commonly prescribed by Chinese medicine physicians to treat breast cancer patients, and our previous study found that its treatment was associated with reduced mortality in these patients. To further explore its therapeutic potential, we investigated the combined effect of its key ingredients, Coptis rhizome and rhubarb, on the metastasis of TNBC by using MDA-MB-231 cell line.

Ethnopharmacological Relevance: Diabetic ulcer is a serious diabetes complication and a primary reason for amputations. For many years, the San Huang Xiao Yan (SHXY) recipe has served as a conventional remedy for these ulcers, effectively reducing inflammatory factors and exhibiting considerable therapeutic efficacy. However, the precise mechanism remains incompletely understood.

Article Synopsis
  • San-Huang-Xie-Xin-Tang (SHXXT) is a traditional Chinese medicine used for treating conditions like hypertension and gastritis, but its chemical composition has been unclear, making it hard to identify effective compounds and ensure quality control.
  • Researchers conducted a detailed analysis of SHXXT's constituents using advanced techniques like HPLC-ESI-QTOF-MS/MS, identifying 45 different compounds, including phenolic acids, flavonoids, alkaloids, anthraquinones, and lignan.
  • This comprehensive study lays the groundwork for future research on the active compounds and mechanisms of action in SHXXT, enhancing our understanding of its efficacy.

Ethnopharmacological Relevance: San Huang Pill (SHP) is a prescription in Dunhuang Ancient Medical Prescription, which has the efficacy of heat-clearing and dampness-drying, and is a traditional formula for the treatment of gastrointestinal diseases. However, its efficacy and mechanism in treating ulcerative colitis (UC) are still unclear.

Aim Of The Study: To investigate the protective effects of SHP and its bioactive compounds against Dextran Sulfate Sodium (DSS)-induced intestinal damage using the Drosophila melanogaster model, and to detect the molecular mechanism of SHP in the treatment of UC.
