Article Abstract

The construction of three-dimensional (3D) covalent organic frameworks (COFs), especially fully conjugated 3D COFs, is a long-standing challenge. Herein, we report a saddle-like, π-conjugated cyclooctatetrathiophene (COTh) as a tetrahedral node to construct fully conjugated 3D COFs. The present work enriches the structural diversities and potential applications of 3D COFs.

This study focuses on designing and developing a novel three-dimensional porphyrinic covalent organic framework (3D-Por-COF) to enhance anticancer sono-photodynamic therapy (SPDT). Leveraging the unique structural advantages of 3D COFs, this work addresses the limitations of traditional 2D-Por-COFs, particularly regarding reactive oxygen species (ROS) production and therapeutic efficacy. The newly developed 3D-Por-COF demonstrated significantly higher ROS generation under combined sonodynamic and photodynamic conditions, leading to an improved therapeutic effect against prostate cancer cells.

The effective delivery of pharmaceuticals to the respiratory tract is significantly influenced by the three-dimensional covalent network structure of mucus and the motility of cilia within the airway surface liquid (ASL). This study investigates the dissolution and absorption of three distinct drugs-Salbutamol sulfate (SAL), Tiotropium bromide (TIO), and Rifampicin (RIF)-in the ASL, focusing on individual particles of each drug with an initial diameter of 5 µm. A three-dimensional numerical model that characterizes mucus as a nonlinear viscoelastic fluid was employed for this analysis.
