CUT&RUN is a recently developed chromatin profiling technique that enables high-resolution chromatin mapping and probing. Herein, we describe our adapted CUT&RUN protocol for transcription factors (TFs). Our protocol outlines all necessary steps for TF profiling including the procedure to obtain proteinA-Mnase, while also outlining the bioinformatic pipeline steps required to process, analyze, and identify novel binding sites and sequences. Due to the small number of cells required, this method will allow the elucidation of cell context-dependent functions of many TFs. For details on the use and execution of this protocol, please refer to Kong et al. (2021).

BackgroundThis investigation aimed to determine the utility of postnatal, ultrasonographically-derived dimensions of the thymus and spleen as imaging indicators for the prediction of early-onset neonatal sepsis (EOS).Material and MethodIn this case-control study, 30 term neonates diagnosed with Early-Onset Sepsis (EOS), based on European Medicines Agency (EMA) criteria, were compared to 30 healthy, matched control neonates. All participants underwent ultrasonography to quantify thymic and splenic dimensions.

The differential diagnosis within polyuria-polydipsia syndrome, especially in the pediatric population, remains challenging. Despite its limited accuracy, the water deprivation test (WDT) is the reference test in pediatrics. We retrospectively analyzed performed in 65 pediatric patients (mean age 8.

Low dose computed tomography (LDCT) imaging obtained for other clinical indications is useful for the opportunistic screening of osteoporosis and demonstrates additional potential for the screening of metabolic risk through the measurement of visceral adipose tissue. In this study, we explored LDCT-derived VAT and calculated VAT thresholds indicative of elevated metabolic risk in a population cohort of Chinese men and women.

Background: Postoperative late recurrence (POLAR) after 2 years from the date of surgical resection of hepatocellular carcinoma (HCC) represents a unique surveillance and management challenge. Despite identified risk factors, individualized prediction tools to guide personalized surveillance strategies for recurrence remain scarce. The current study sought to develop a predictive model for late recurrence among patients undergoing HCC resection.

Adulterated yohimbine (YHB) in food poses a risk to public health, making it imperative to develop fast and sensitive detection methods. In this study, computational-chemistry-based prediction was employed to design YHB haptens for generating the high-affinity monoclonal antibody Yohi-4A7, which exhibited an optimal half-inhibitory concentration (IC) of 1.69 ng/mL against YHB.
