Background And Purpose: Amyotrophic lateral sclerosis (ALS) patients exhibit dysfunctional energy metabolism and weight loss, which is negatively correlated with survival, together with neuroinflammation. However, the possible contribution of neuroinflammation to deregulations of feeding behaviour in ALS has not been studied in detail. We here investigated if microglial K 3.1 is linked to hypothalamic neuroinflammation and affects feeding behaviours in ALS mouse models.
Experimental Approach: hSOD1 and TDP43 mice were treated daily with 120 mg·kg of TRAM-34 or vehicle by intraperitoneal injection from the presymptomatic until the disease onset phase. Body weight and food intake were measured weekly. The later by weighing food provided minus that left in the cage. RT-PCR and immunofluorescence analysis were used to characterize microglia phenotype and the main populations of melanocortin neurons in the hypothalamus of hSOD1 and age-matched non-tg mice. The cannabinoid-opioid interactions in feeding behaviour of hSOD1 mice were studied using an inverse agonist and an antagonist of the cannabinoid receptor CB (rimonabant) and μ-opioid receptors (naloxone), respectively.
Key Results: We found that treatment of hSOD1 mice with the K 3.1 inhibitor TRAM-34 (i), attenuates the pro-inflammatory phenotype of hypothalamic microglia, (ii) increases food intake and promotes weight gain, (iii) increases the number of healthy pro-opiomelanocortin (POMC) neurons and (iv), changes the expression of cannabinoid receptors involved in energy homeostasis.
Conclusion And Implications: Using ALS mouse models, we describe defects in the hypothalamic melanocortin system that affect appetite control. These results reveal a new regulatory role for K 3.1 to counteract weight loss in ALS.

The brown planthopper (BPH), Nilaparvata lugens (Stål), is a typical insecticide-induced resurgence rice pest that causes severe damage to rice in Asian countries. Previous studies have shown that the fungicide Jinggangmycin (JGM), used to control rice sheath blight disease, can stimulate BPH fecundity; however, the molecular mechanism remains to be further explored. In this study, based on transcriptomic analysis, we found that the PI3K-Akt signaling pathway was significantly enriched in BPH after feeding on JGM-treated rice, where the NlPR-L and NlABD4-L genes were significantly upregulated.

In live specimens of the nemertodermatidan Flagellophora apelti Faubel and Dörjes, 1978, a peculiar organ looking like a fascicle of bristles-and so called a broom organ by its discoverer-occupies the front third or so of the body. The animal can extrude the organ to splay the bristles in a fan-like array, each bristle having an adhesive tip. Described first by light histology as a bundle of flagella, this organ can be seen by transmission electron microscopy to be actually a bundle of exceedingly long necks of glands.

The objective of this experiment was to evaluate the effects of dietary starch concentration by replacing high-moisture corn with soy hulls and a direct-fed microbial (DFM) containing Bacillus subtilis 810 and Bacillus licheniformis 809 on the lactation performance, feeding behavior, total-tract nutrient digestibility, and enteric CH emissions of dairy cows. Sixty-four multiparous Holstein cows (67 ± 24 DIM and 729 ± 60 kg of BW at the beginning of the covariate period) were randomly assigned to 1 of 4 treatments with a 2 × 2 factorial arrangement for a 12-wk treatment period, after a 1-wk acclimation period and 2-wk covariate period. Treatments consisted of diets with 21% starch and no DFM (RS-CON), 21% starch and DFM (RS-DFM), 27% starch and no DFM (NS-CON), and 27% starch and DFM (NS-DFM).
