Traditional live attenuated vaccines (LAVs) are typically developed through serial passaging or genetic engineering to introduce specific mutations or deletions. While viral RNA secondary or tertiary structures have been well-documented for their multiple functions, including binding with specific host proteins, their potential for LAV design remains largely unexplored. Herein, using Zika virus (ZIKV) as a model, we demonstrate that targeted disruption of the primary sequence or tertiary structure of a specific viral RNA element responsible for Musashi-1 (MSI1) binding leads to a tissue-specific attenuation phenotype in multiple animal models.

The NS5 RNA-dependent RNA polymerase (RdRp) of dengue virus (DENV) and Zika virus (ZIKV) is highly conserved, making nucleoside-based RdRp inhibitors a promising strategy for antiviral development. While nucleoside analogs have shown strong clinical potential, challenges such as cell permeability, the efficiency of triphosphate conversion, degradation, and mitochondrial toxicity remain.

Zika virus (ZIKV) has emerged as a rising concern in global health in recent years. The role of PD-1/PD-L1 immune checkpoint in acute ZIKV infection remains to be understood. In this study we demonstrated the activation of PD-1/PD-L1 immune checkpoint by ZIKV.

Intracranial calcifications (ICCs) are a characteristic neuropathological feature of several congenital viral infections, including Zika virus (ZIKV), cytomegalovirus (CMV), and lymphocytic choriomeningitis virus (LCMV). These lesions are linked to severe neurodevelopmental outcomes, such as microcephaly, epilepsy, and cognitive deficits, yet the mechanisms underlying their formation and resolution remain unclear. ICCs are thought to arise from an imbalance in osteogenic and osteolytic signaling in the developing brain.

Background: Dengue, chikungunya, and Zika present significant public health challenges in Colombia. Spatial studies help clarify the distribution and progression of these diseases over time and location. Objective to describe the spatio-temporal distribution and clustering patterns of dengue, chikungunya, and Zika in Medellín, Colombia, between 2013 and 2021, with the aim of providing baseline spatial intelligence to support future epidemiological and policy-oriented analyses.
