Article Abstract

Objectives: We tested if METCAM/MUC18 overexpression also plays a suppressor role in another human ovarian cancer cell line, BG-1, in addition to the SK-OV3 cell line.

Methods: Human ovarian cancer BG-1 cells were transfected with METCAM/MUC18 cDNA and G418-resistant clones expressing different levels of METCAM/MUC18 were isolated. These clones were used to test the effects of enforced expression of METCAM/MUC18 on in vitro motility, invasiveness, and anchorage-independent colony formation (in vitro tumorigenesis), and in vivo tumorigenesis after SC injection and after IP injection in female athymic nude mice.

Results: Overexpression of METCAM/MUC18 reduced in vitro motility and invasiveness of BG-1 cells and anchorage-independent colony formation (in vitro tumor formation). Higher expression of METCAM/MUC18 in BG-1 cells significantly reduced in vivo tumor proliferation of the BG-1 cells after IP injection (orthotopic route) of the clones in female nude mice, though it did not significantly affect in vivo tumor proliferation after SC injection (non-orthotopic route).

Conclusion: Similar to SK-OV3 cells, METCAM/MUC18 also plays a suppressor role in the progression of BG-1 cells in a xenograft mouse model.

The study aims to design and fabricate an ultra-easier multi-functional biomedical polymeric scaffold loaded with unique equimolar Ca:P phasic bioactive glass material (BG). Gelatin (G) - 45S5 bioactive glass (BG) scaffolds were synthesized via a simple laboratory refrigerator with higher biocompatibility and cytocompatibility. The results proved that BG has enhanced bio-mineralization of the scaffolds and results support that the G: BG (1:2) ratio is the more appropriate composition.

Red blood cell parameters were assessed in a natural population of the northern red-backed vole (Clethrionomys rutilus Pallas, 1779) in the zone of influence of the Kirovgrad Copper Smelter along a gradient of pollution by heavy metals (Cu, Zn, Cd, and Pb) at three catching sites (polluted [Imp] and controls [Bg-1, and Bg-2]). The difference of the smelter area (Imp group of voles) from both background groups (Bg-1 and Bg-2) was proven by means of a set of 13 parameters in univariate and multivariate analyses. Among the detected erythrocyte disturbances, we noted the following: a decrease in activities of Na,K-ATPase and antioxidant enzymes (SOD, GSH-Px, and CAT); an increase in the concentration of lipid peroxidation products, in osmotic fragility, and in intravascular hemolysis; interruption of carbohydrate metabolism; and lowered oxygen-carrying capacity.

Objective: To investigate the molecular mechanism of the proliferation and migration of BG-1 and MCF-7cells induced by DEHP, and the antagonistic effect of metformin.

Methods: The proliferation, cell cycle progression, migration, and invasion abilities of BG-1 and MCF-7 cancer cells were examined Cell Counting Kit-8, flow cytometry, Transwell, and scratch assays. E2F1, SKP2, cyclin D1, vimentin, E-cadherin, and GSK-3β, all of which play key roles in cancer development the PI3K/AKT signaling pathway, were examined by immunofluorescence and immunocytochemistry.
