Article Abstract

Distal enhancers play pivotal roles in development and disease yet remain one of the least understood regulatory elements. We used massively parallel reporter assays to perform functional comparisons of two leading enhancer models and find that gene-distal transcription start sites are robust predictors of active enhancers with higher resolution than histone modifications. We show that active enhancer units are precisely delineated by active transcription start sites, validate that these boundaries are sufficient for capturing enhancer function, and confirm that core promoter sequences are necessary for this activity. We assay adjacent enhancers and find that their joint activity is often driven by the stronger unit within the cluster. Finally, we validate these results through functional dissection of a distal enhancer cluster using CRISPR-Cas9 deletions. In summary, definition of high-resolution enhancer boundaries enables deconvolution of complex regulatory loci into modular units.

Developing artificial hosts with temperature-driven conformational switching behaviors facilitates our understanding of the temperature-dependent allostery and adaptation mechanisms in natural recognition systems. Herein, we report the design and synthesis of three pairs of water-soluble, enantiomeric binaphthalene-based tetraimidazolium macrocycles (SS/RR-1•4Cl- - SS/RR-3•4Cl-) as artificial hosts for exploring sequence-selective recognition of dinucleotides in aqueous media. Owing to the reversible rotational conformation of axially chiral binaphthyl units, SS-1•4Cl- demonstrates the conformational switching, converting from cis-conformation (SS-1) to trans-conformation (SS-1) by increasing temperature, thereby causing the recognition cavity to transition from a closed to an open state.

Strong intermolecular interactions facilitate the formation of efficient ion transport channels, which, in turn, significantly boost the performance of anion exchange membranes (AEMs). Herein, 9-anthracene methanol with both π-π stacking and hydrogen bonding intermolecular forces is used as a bifunctional unit to synthesize high-performance AEMs through the Friedel-Crafts superacid catalytic reaction for the first time. The π-π stacking in the bifunctional units can induce hydrophilic pyridine cations to aggregate, and the hydrogen bonding can provide transport sites for OH and water molecules in the hydrophobic component.
