The APEX2 gene encodes APE2, a nuclease related to APE1, the apurinic/apyrimidinic endonuclease acting in base excision repair. Loss of APE2 is lethal in cells with mutated BRCA1 or BRCA2, making APE2 a prime target for homologous recombination-defective cancers. However, because the function of APE2 in DNA repair is poorly understood, it is unclear why BRCA-deficient cells require APE2 for viability. Here we present the genetic interaction profiles of APE2, APE1, and TDP1 deficiency coupled to biochemical and structural dissection of APE2. We conclude that the main role of APE2 is to reverse blocked 3' DNA ends, problematic lesions that preclude DNA synthesis. Our work also suggests that TOP1 processing of genomic ribonucleotides is the main source of 3'-blocking lesions relevant to APEX2-BRCA1/2 synthetic lethality. The exquisite sensitivity of BRCA-deficient cells to 3' blocks indicates that they represent a tractable vulnerability in homologous recombination-deficient tumor cells.

Characteristic boundary conditions are widely used to simulate compressible flow-induced noise. This study extends the application to incompressible flow-induced noise by introducing a low Mach number characteristic boundary condition (LMCBC) to eliminate sound wave reflections at boundaries. This boundary condition is derived from the second type of acoustic perturbation equations (APE-2) and implemented within the open-source software OpenFOAM (OpenCFD Ltd, Bracknell, Berkshire, UK).

Introduction: Within the spectrum of epilepsies, autoimmune-associated epilepsy (AAE) reflects the complexity of its presentations including refractory seizures along with cognitive decline and behavioral or psychiatric dysfunction. The suspicion of autoimmune epilepsy is heightened in individuals who have a pre-existing autoimmune condition, such as type 1 diabetes mellitus (T1DM). This report details a case of autoimmune epilepsy associated with Glutamic acid decarboxylase (GAD65) antibodies coexisting with T1DM.

Unlabelled: Cisplatin remains a cornerstone chemotherapy for many solid tumors but is limited by dose-limiting toxicities, including nephrotoxicity, peripheral neuropathy, and ototoxicity-the latter of which disproportionately affects pediatric patients and lacks effective prevention strategies. Although therapeutic approaches to mitigate cisplatin-induced toxicity are urgently needed, the underlying mechanisms driving organ-specific injury remain incompletely understood. Background: To investigate antibody prevalence and long-term seizure outcomes in epilepsy patients with suspected autoimmune etiology but without any signs of autoimmune encephalitis (AE), and to compare the performance of Antibody Prevalence in Epilepsy and Encephalopathy (APE2) score and Antibodies Contributing to Focal Epilepsy Signs and Symptoms (ACES) score.
Methods: 123 patients diagnosed with epilepsy with possible autoimmune etiology but without any signs of AE at the Second Xiangya Hospital from January 2016 to July 2023 were included, We compared clinical and paraclinical findings in included patients and analyzed the factors influencing long-term seizure outcomes. The APE2 and ACES scores were performed by external validation and sensitivity and specificity analysis.
