Article Abstract

The purpose of the present research was to develop a suitable, simple, precise, accurate, robust, and reproducible RP-HPLC method for a reliable simultaneous quantification of docetaxel (DTX) and curcumin (CCM) in rat plasma samples using paclitaxel (PTX) as an internal standard. The samples were assayed by the Agilent 1260 Infinity HPLC instrument using a Capcell Pak C column (4.6 mm × 150 mm, 5 µm) under isocratic conditions. The mobile phase consisted of acetonitrile and triple distilled water (40/60, v/v) with a flow rate of 1.0 ml/min. The eluent was monitored at 230 nm for simultaneous measurement of curcumin and docetaxel. The method was validated by determining system suitability, selectivity, sensitivity, linearity, inter-day and intra-day precision, accuracy, robustness, and stability in accordance with the guidelines of the United States Food and Drug Administration (FDA). The developed chromatographic method proved to be simple, precise, accurate, robust and reproducible. Moreover, the samples showed stability at room temperature over a period of 48 h. Thus, this method would be employed for routine simultaneous quantification of docetaxel and curcumin in rat plasma samples.

Background: Astaxanthin Z-isomers have attracted much attention because recent studies have demonstrated that they exhibit greater bioavailability and biological activity than the naturally predominant all-E-isomer. However, the plasma appearance and tissue distribution of astaxanthin isomers when administered with a diet rich in astaxanthin Z-isomers are largely unknown. To understand the health benefits and safety of astaxanthin Z-isomers, it is important to study the in vivo kinetics of the isomers.

Famotidine (FMD) is an H₂-receptor antagonist with limited oral bioavailability and a short plasma half-life (2.5-4 h). Silk fibroin-chitosan nanoparticles (FBN-CS-NPs) represent a novel nanocarrier approach for treating peptic ulcers, combining biocompatibility, mucoadhesiveness, and pH-sensitive release.

Aims: Acute restraint stress (RS) has been reported to activate the supraoptic nucleus of the hypothalamus (SON). The aim of the present study was to evaluate the role of glutamatergic neurotransmission in the SON on autonomic [mean arterial pressure (MAP), heart rate (HR), and tail cutaneous temperature], neuroendocrine (plasma levels of corticosterone, oxytocin, and vasopressin), and behavioral responses to RS.

Methods: Male Wistar rats with bilateral SON cannulas received microinjections of NMDA or non-NMDA receptor antagonists or vehicle before restraint stress, and the effects on cardiovascular, tail temperature, hormonal, and behavioral responses were evaluated RESULTS: Microinjection of DL-AP7 or NBQX into the SON reduced MAP increases and tail temperature decreases induced by RS.

Both the autonomic nervous system and the hypothalamic-pituitary-adrenal axis respond to systemic immune challenge by initiating anti-inflammatory reflexes. Here we compare those two homeostatic responses in vivo. We first confirmed in male urethane-anaesthetized rats that disabling the autonomic reflex by bilateral section of the splanchnic sympathetic nerves increased plasma tumor necrosis factor α (TNF) responses to systemic lipopolysaccharide (LPS, 60 µg/kg i.

The occurrence of isomeric dihydroflavone and chalcone, two important subfamilies of flavonoid class, extensively happens in herbal medicines. However, identical MS/MS spectra make the identity confirmation a tough job, the complexity will be further boosted in biological samples. Inspired by that isomers possess distinct inherent physicochemical parameters, optimal collision energy (OCE), which is positively correlated with the bond dissociation energies (BDEs), was evaluated towards differentiating isomeric dihydroflavone and chalcone.
