Article Abstract

Reduced glutathione (GSH) level inside the cell is a critical determinant for cell viability. The level of GSH varies across the cells, tissues and environmental conditions. However, our current understanding of physiological and pathological GSH changes at high spatial and temporal resolution is limited due to non-availability of practicable GSH-detection methods. In order to measure GSH at real-time, a ratiometric genetically encoded nanosensor was developed using fluorescent proteins and fluorescence resonance energy transfer (FRET) approach. The construction of the sensor involved the introduction of GSH binding protein (YliB) as a sensory domain between cyan fluorescent protein (CFP; FRET donor) and yellow fluorescent protein (YFP; FRET acceptor). The developed sensor, named as FLIP-G (Fluorescence Indicator Protein for Glutathione) was able to measure the GSH level under in vitro and in vivo conditions. When the purified FLIP-G was titrated with different concentrations of GSH, the FRET ratio increased with increase in GSH-concentration. The sensor was found to be specific for GSH and also stable to changes in pH. Moreover, in live bacterial cells, the constructed sensor enabled the real-time quantification of cytosolic GSH that is controlled by the oxidative stress level. When expressed in yeast cells, FRET ratio increased with the external supply of GSH to living cells. Therefore, as a valuable tool, the developed FLIP-G can monitor GSH level in living cells and also help in gaining new insights into GSH metabolism.

Form and function in biological filaments: a physicist's review.

Nature uses elongated shapes and filaments to build stable structures, generate motion and allow complex geometric interactions. In this review, we examine the role of biological filaments across different length scales. From the molecular scale, where cytoskeletal filaments provide a robust but dynamic cellular scaffolding, over the scale of cellular appendages like cilia and flagella, to the scale of filamentous microorganisms like cyanobacteria, among the most successful genera on Earth, and even to the scale of elongated animals like worms and snakes, whose motility modes inspire robotic analogues.

Flagellates, unicellular organisms equipped with one or a few flagella, are phylogenetically and functionally hugely diverse. Yet, most studies have focused on a few model organisms and on the role of the flagellum in propulsion, ignoring the fundamental role of the flagellum in foraging. The number and position of flagella vary between species; the flagella may be naked or equipped with vanes or hairs; the kinematics and wave patterns vary and may be planar or three-dimensional; and the flagellum may extend from the surface of the cell or lie within a groove on the cell surface.

The donor/acceptor (D/A) interfaces in bulk heterojunction (BHJ) organic solar cells (OSCs) critically govern exciton dissociation and molecular diffusion, determining both efficiency and stability. Herein, we design a double-cable conjugated polymer, SC-1F, to insert into a physically-blended D/A system to optimize the interface. We have found that SC-1F spontaneously segregates to the interface through favorable miscibility and heterogeneous nucleation with the acceptor.

However, how TCF-1 is downregulated remains unclear.
