Human fungal infections comprise systemic mycoses as well as various skin diseases. Rising case numbers along with inefficient therapies and the appearance of drug-resistant strains unleashed a considerable health problem over the last years. Thus, the identification and development of new antifungal drugs is mandatory, which can include the design of new antifungals, or, more time saving, the repurposing of known drugs already applied for the therapy of other human diseases.

While there are currently no approved HEV-specific therapeutics, ribavirin and pegylated interferon, prescribed off-label, are the current standard of care. However, ribavirin resistance and toxicity highlight the unmet clinical need to identify safer, HEV-specific antivirals.

Background: Variants of SORL1 have been associated with both late and early onset of Alzheimer's disease (AD). SORL1 encodes the sorting-related receptor with A repeat (SORLA) protein, which belongs to the VPS10 receptor family. SORLA protects against AD pathogenesis through its sorting function, and reduced SORLA levels have been consistently observed in sporadic AD.

Background: Candida auris is a significant clinical concern due to its ability to cause outbreaks in healthcare settings and its common resistance to current treatments. This highlights the need for alternative therapies. Drug repurposing offers a promising approach, and the combination of pentamidine (antiprotozoal) and auranofin (anti-rheumatic) has shown potential antifungal activity against Candida species, including C.

Although numerous drugs have been tested to treat ovarian cancer (OC), survival rates remain low as there has been no major improvement from platinum (Pt)-based therapy and there is a high rate of Pt resistance in these tumors. Following several rounds of chemotherapy, OC cells develop Pt-resistance by increasing DNA repair and antioxidant defense mechanisms. This study aimed to design a treatment to combat recurrent stages of OC by repurposing the anti-rheumatic gold complex auranofin (AF).
