Traditional Chinese Medicines are promising sources of new agents for controlling cancer metastasis. Compound Kushen Injection (CKI), prepared from medicinal plants and , disrupts cell cycle and induces apoptosis in breast cancer; however, effects on migration and invasion remained unknown. CKI, fractionated mixtures, and isolated components were tested in migration assays with colon (HT-29, SW-480, DLD-1), brain (U87-MG, U251-MG), and breast (MDA-MB-231) cancer cell lines. Human embryonic kidney (HEK-293) and human foreskin fibroblast (HFF) served as non-cancerous controls. Wound closure, transwell invasion, and live cell imaging showed CKI reduced motility in all eight lines. Fractionation and reconstitution of CKI demonstrated combinations of compounds were required for activity. Live cell imaging confirmed CKI strongly reduced migration of HT-29 and MDA-MB-231 cells, moderately slowed brain cancer cells, and had a small effect on HEK-293. CKI uniformly blocked invasiveness through extracellular matrix. Apoptosis was increased by CKI in breast cancer but not in non-cancerous lines. Cell viability was unaffected by CKI in all cell lines. Transcriptomic analyses of MDA-MB-231indicated down-regulation of actin cytoskeletal and focal adhesion genes with CKI treatment, consistent with observed impairment of cell migration. The pharmacological complexity of CKI is important for effective blockade of cancer migration and invasion.

Colorectal cancer (CRC) remains one of the most lethal malignancies globally, driven by complex molecular mechanisms that contribute to its progression and metastasis. This study focuses on the role of N1-methyladenosine (mA) RNA methylation in CRC, particularly its effect on Rab Interacting Lysosomal Protein-Like 1 (RILPL1) expression and the downstream activation of the CaMKII/CREB signaling pathway. Bioinformatics analysis identified RILPL1 as a key gene associated with poor CRC prognosis, exhibiting increased expression levels in cancerous tissues, with further elevation in metastatic samples.

Glioblastoma multiforme (GBM) is a rapidly progressing brain malignancy, with its progression closely tied to a hypoxic microenvironment. Hypoxia-inducible factor-1α (HIF-1α) acts as a vital regulator in tumor adaptation to low oxygen levels, and its relationship with the Wnt/β-catenin signaling pathway exerts significant functions in the malignant properties of GBM. Ring finger protein 180 (RNF180) is an E3 ubiquitin-protein ligase that promotes polyubiquitination and degradation. We analyzed the roles and molecular mechanisms of RNF180 during the tumorigenesis and progression of colorectal cancer (CRC) through bioinformatics analysis, in vivo and vitro experiments. Infected wounds remain a major clinical challenge due to bacterial invasion, which disrupts the natural healing cascade through excessive reactive oxygen species (ROS) generation, severe vascular damage, and persistent inflammation. Inspired by the catechol-rich adhesive domains of mussel foot proteins, we developed an extracellular matrix (ECM)-mimetic polyethylene glycol (PEG) hydrogel incorporating polydopamine (PDA)-functionalized zinc oxide nanoparticles (ZnONPs) for infected wound therapy. The amino acid-functionalized PEG hydrogel reproduces ECM-like properties to facilitate cell migration and efficient exudate management; however, its lack of intrinsic antimicrobial activity limits therapeutic efficacy.
