Platelet Rich Plasma (PRP) has been utilized to augment ACL surgery, with studies reaching conflicting conclusions as to the extent of beneficial effects. While the majority of ACL surgeries I have performed have not been augmented, I have utilized PRP (and bone marrow concentrate) enough to conclude that my personal experience matches literature finding no significant difference in outcomes, and, at present does not support routine augmentation of ACL reconstruction (ACLR) with PRP. Instead, I believe the "crimson duvet" effect of reaming tunnels is a key source of cells and signals that promote healing and initiate graft incorporation.

Cellular Immunogenicity Assessments in CAR-T Cell Therapies: Current Insights and Future Directions.

AAPS J

September 2025

Precision Medicine Bioanalytical & Translational Sciences, Bristol Myers Squibb, Route 206 & Province Line Road, Princeton, NJ, 08543, USA.

CAR-T-cells can drive MHC class-I-mediated CD8 + cytotoxic T-cell response towards CAR constructs in addition to an antibody response. Immune response may also develop towards residuals present in the CAR-T cell product such as AAV, CRISPR/CAS9, and expamers. Health authorities recommend developing assays to assess both humoral and cellular immunogenicity towards the CAR-T protein.

Aims: Calcific aortic valve disease is the most common valvular heart disease characterized by an inflammatory response in the leaflets followed by fibro-calcific remodelling of valvular interstitial cells (VICs). Lipoprotein(a) [Lp(a)] is a well-recognized risk factor for CAVD, however the role of metabolism in driving Lp(a)-induced inflammation remains largely elusive. Therefore, we aim to investigate the role of Lp(a) in driving inflammatory and metabolic changes in VICs and examine how alterations in cellular metabolism can alter their inflammatory phenotype.

Polyploid giant cancer cells: Unveiling a latent axis of resistance and tumor evolution in HER2-targeted therapy.

Cancer Lett

Recent findings by Yazdi et al. in Cancer Letters present a mechanistic breakthrough in understanding resistance to HER2-targeting antibody-drug conjugates (ADCs). Their identification of drug-persistent polyploid giant cancer cells (PGCCs) as active participants in resistance redefines our understanding of intratumoral plasticity and therapeutic evasion.
