Many implementations of pooled screens in mammalian cells rely on linking an element of interest to a barcode, with the latter subsequently quantitated by next generation sequencing. However, substantial uncoupling between these paired elements during lentiviral production has been reported, especially as the distance between elements increases. We detail that PCR amplification is another major source of uncoupling, and becomes more pronounced with increased amounts of DNA template molecules and PCR cycles. To lessen uncoupling in systems that use paired elements for detection, we recommend minimizing the distance between elements, using low and equal template DNA inputs for plasmid and genomic DNA during PCR, and minimizing the number of PCR cycles. We also present a vector design for conducting combinatorial CRISPR screens that enables accurate barcode-based detection with a single short sequencing read and minimal uncoupling.

The objective of this study was to evaluate the concentration and integrity index of circulating cell-free DNA (ccf-DNA) as biomarkers for the detection and monitoring of minimal residual disease (MRD) in pediatric patients with B-cell acute lymphoblastic leukemia (B-ALL). Comparison with a validated methodology for the quantification of monoclonal rearrangements of the IGH gene was made. Background: Nucleic Acid Amplification Tests (NAAT) remain one of the most reliable methods for pathogen identification. Given the high false-negative rates associated with traditional staining and microscopic examination, the time-consuming nature and low sensitivity of bacterial culture methods, as well as the inability of conventional NAAT to achieve absolute quantification.
Methods: To achieve rapid and quantitative detection of , we selected the 23S rRNA gene as the target for identification and developed a droplet digital PCR detection method.

Introduction: Acute coronary syndrome (ACS) is a leading cause of death, and clopidogrel resistance remains a major challenge in its treatment. This study aims to determine the impact of CYP2C19 genetic variants on clopidogrel resistance (CR) and major adverse cardiovascular events (MACEs) in Vietnamese patients undergoing percutaneous coronary intervention (PCI).
Methods: We carried out a descriptive cross-sectional study, supplemented by a prospective longitudinal follow-up, on 113 ACS patients undergoing PCI with drug-eluting stent implantation at the Department of Cardiology, Military Hospital 103, from January 2015 to May 2018.
