Article Abstract

The pH labile metabolite, hydrophobicity, high oral dose and systematic exposure of GDC-0810 posed tremendous challenges to develop a LC-MS method for a stable isotope labeled aBA study. In this study, we explored practical solutions to balance stability and sensitivity and to cope with the impact of high C to C ratio on the labeling selection and assay dynamic range. A [C] GDC-0810 was synthesized to minimize the isotopic interference between PO dose, internal standard and I.V. microtracer. A highly sensitive LC-MS assay was validated for quantitation of [C] GDC-0810 from 5 to 1250 pg/mL. The optimized method was applied to a proof of concept cynomolgus monkey aBA study and the bioavailability calculated using microtracer dosing and regular dosing were similar to each other.

Karst water bodies are vital groundwater resources particularly vulnerable to pollution. Protecting their water quality requires documenting contaminants traditionally associated with anthropogenic activities (metals, nutrients, and fecal indicator bacteria) as well as emerging contaminants, such as antibiotic-resistant organisms (AROs) and perfluoroalkyl substances (PFAS). This study detected contaminants in karst-associated water bodies on the Yucatán Peninsula, including 10 sinkholes (cenotes) and one submarine groundwater discharge (SGD) site.

Animal migration remains poorly understood for many organisms, impeding understanding of movement dynamics and limiting conservation actions. We develop a framework that scales from movements of individuals to the dynamics of continental migration using data synthesis of endogenous markers, which we apply to three North American bat species with unexplained high rates of fatalities at wind energy facilities. The two species experiencing the highest fatality rates exhibit a "pell-mell" migration strategy in which individuals move from summer habitats in multiple directions, both to higher and lower latitudes, during autumn.
