Background: KRAS mutations are typically mutually exclusive in non-small cell lung cancer (NSCLC), with the G12C mutation being the most common subtype. The coexistence of KRAS and EGFR mutations is exceedingly rare and is typically emerges as a secondary event following acquired resistance to EGFR-targeted therapies. We presented a case of a newly diagnosed NSCLC patient harboring concurrent EGFR L858R and KRAS G12A mutations.

Diagnostic value of F-FDG PET/CT in the follow-up of conjunctival melanoma.

Objective: To evaluate the diagnostic performance of F-fluorodeoxyglucose positron emission tomography/computed tomography (F-FDG PET/CT) in detecting metastatic conjunctival melanoma (CM).

Methods: This retrospective study enrolled 67 patients with histopathologically confirmed CM who underwent F-FDG PET/CT for follow-up or suspected recurrence. Parameters including short-axis diameter of lesions, maximum standardized uptake value (SUVmax), the target-to-nontarget (T/NT) ratios of cervical lymph nodes, and bone lesion characterization (osteolytic, osteoblastic, or unchanged) were evaluated.

The management of medulloblastoma (MB) remains a significant challenge, primarily attributed to the presence of cancer stem cells and the inadequate delivery of therapeutic agents across the blood-brain barrier. GLI, as a regulator of the hedgehog signaling pathway in normal cerebellum development, also exerts pivotal functions in MB initiation, progression, and metastasis and maintains the stemness of MB stem cells. In this study, we devised a combined therapeutic approach by integrating the BRD4 inhibitor JQ1 with the SMO inhibitor saikosaponin B1 (SSB1) to inhibit MB via regulation of GLI activation.
