Article Abstract

We propose a method to improve the resolution of coherent anti-Stokes Raman scattering microscopy (CARS), and present a theoretical model. The proposed method, coherent anti-Stokes Raman scattering difference microscopy (CARS-D), is based on the intensity difference between two differently acquired images. One being the conventional CARS image, and the other obtained when the sample is illuminated by a doughnut shaped spot. The final super-resolution CARS-D image is constructed by intensity subtraction of these two images. However, there is a subtractive factor between them, and the theoretical model sets this factor to obtain the best imaging effect.

Multimodal imaging techniques have the potential to enhance the interpretation of histology by offering additional molecular and structural information beyond that accessible through hematoxylin and eosin (H&E) staining alone. Here, we present a quantitative approach for comparing the information content of different image modalities, such as H&E and multimodal imaging. We used a combination of deep learning and radiomics-based feature extraction with different information markers, implemented in Python 3.

We performed ultrafast reflection-type pump-probe spectroscopy of coherent longitudinal optical (LO) phonons in a (001)-oriented semi-insulating GaAs crystal using femtosecond laser pulses optically transparent to GaAs. From the temporal oscillatory profile as a function of time delay, the decay time of the coherent LO phonons was estimated to be (4.4±0.

A technique for quantitative two-photon laser-induced fluorescence measurements of the ←(=0) Hopfield-Birge system in high-enthalpy environments is shown. The two-photon transition is pumped by high-intensity, nanosecond laser excitation to produce photoionization rates much greater than the quenching rates at the probe volume, which simplifies the quantification of the signal. We demonstrate this technique along the stagnation streamlines of a graphite ablator exposed to the 6000 K plume of an atmospheric-pressure inductively coupled air plasma torch.

A novel, to our knowledge, counter-propagating (CoP) phase-matching configuration of hybrid femtosecond/picosecond coherent anti-Stokes Raman scattering (fs/ps CARS) for ultra-high-resolution measurements of gas temperature is demonstrated. The resulting spatial resolution is on the order of 10s of µm-more than an order of magnitude improvement over traditional phase-matching geometries-which enables spatially resolved gas-phase measurements of steep temperature gradients and minimizes effects of spatial averaging. Two additional advantages of this measurement system are the ability to simply control the location of the measurement volume by adjusting relative pulse timing and the size of the probe volume by adjusting pulse duration.

The multimodal nonlinear optical imaging offers simultaneous multi-component analysis for biological tissues. In this work, large-region multimodal nonlinear optical imaging with a low aberration mosaic method is demonstrated. A multimodal nonlinear optical imaging platform capable of simultaneous two-photon excitation fluorescence (TPEF) and spectral focusing coherent anti-Stokes Raman scattering (SF-CARS) imaging is built.
