Considering the biological roles of WNT4 and WNT5A involved in adipogenesis, we aimed to investigate whether SNPs in WNT4 and WNT5A contribute to obesity related traits in Han Chinese population. Targeted genomic sequence for WNT4 and WNT5A was determined in 100 Han Chinese subjects and tag SNPs were selected. Both single SNP and SNP × SNP interaction association analyses with body mass index (BMI) were evaluated in the 100 subjects and another independent sample of 1,627 Han Chinese subjects. Meta-analyses were performed and multiple testing corrections were carried out using the Bonferroni method. Consistent with the Genetic Investigation of ANthropometric Traits (GIANT) dataset results, we didn't detect significant association signals in single SNP association analyses. However, the interaction between rs2072920 and rs11918967, was associated with BMI after multiple testing corrections (combined P = 2.20 × 10). The signal was also significant in each contributing data set. SNP rs2072920 is located in the 3'-UTR of WNT4 and SNP rs11918967 is located in the intron of WNT5A. Functional annotation results revealed that both SNPs might be involved in transcriptional regulation of gene expression. Our results suggest that a combined effect of SNPs via WNT4-WNT5A interaction may affect the variation of BMI in Han Chinese population.

Background: Postoperative late recurrence (POLAR) after 2 years from the date of surgical resection of hepatocellular carcinoma (HCC) represents a unique surveillance and management challenge. Despite identified risk factors, individualized prediction tools to guide personalized surveillance strategies for recurrence remain scarce. The current study sought to develop a predictive model for late recurrence among patients undergoing HCC resection.

Rationale: Genome-wide association studies (GWASs) are used to identify genetic variants for association with schizophrenia (SCZ) risk; however, each GWAS can only reveal a small fraction of this association.
Objectives: This study systematically analyzed multiple GWAS data sets to identify gene subnetwork and pathways associated with SCZ.
Methods: We identified gene subnetwork using dmGWAS program by combining SCZ GWASs and a human interaction network, performed gene-set analysis to test the association of gene subnetwork with clinical symptom scores and disease state, meanwhile, conducted spatiotemporal and tissue-specific expression patterns and cell-type-specific analysis of genes in the subnetwork.

Background: As a common postoperative neurological complication, postoperative delirium (POD) can lead to poor postoperative recovery in patients, prolonged hospitalization, and even increased mortality. However, POD's mechanism remains undefined and there are no reliable molecular markers of POD to date. The present work examined the associations of cerebrospinal fluid (CSF) sTREM2 with CSF POD biomarkers, and investigated whether the effects of CSF sTREM2 on POD were modulated by the core pathological indexes of POD (Aβ42, tau, and ptau).

Sonazoid, a combined blood pool and Kupffer-cell agent, can be specifically phagocytosed by Kupffer cells in the liver, allowing lesion detection and characterization of focal liver lesions (FLLs) at the post-vascular phase apart from the vascular phase which is similar to that of other second-generation US contrast agents. Sonazoid CEUS is currently approved for use in some Asian countries. With the increasing use of Sonazoid CEUS for FLLs in clinical practice, developing consensus or guidelines to help standardize its use is required.

Purpose: To explore the causal links between antihypertension drugs usage and age-related macular degeneration (AMD).
Methods: Multiple genetic analyses, including summary data-based Mendelian randomization (SMR), traditional MR, and colocalization analysis, were used to explore the causal associations between antihypertension drugs and AMD. Clinical data from the UK Biobank and the National Health and Nutrition Examination Survey (NHANES) was applied to refined risk assessment of specific antihypertensive medications in the context of AMD development.
