Bone tissue engineering is sought to apply strategies for bone defects healing without limitations and short-comings of using either bone autografts or allografts and xenografts. The aim of this study was to fabricate a thin layer poly(lactic-co-glycolic) acid (PLGA) coated beta-tricalcium phosphate (β-TCP) scaffold with sustained release of vascular endothelial growth factor (VEGF). PLGA coating increased compressive strength of the β-TCP scaffolds significantly. For in vitro evaluations, canine mesenchymal stem cells (cMSCs) and canine endothelial progenitor cells (cEPCs) were isolated and characterized. Cell proliferation and attachment were demonstrated and the rate of cells proliferation on the VEGF released scaffold was significantly more than compared to the scaffolds with no VEGF loading. A significant increase in expression of COL1 and RUNX2 was indicated in the scaffolds loaded with VEGF and MSCs compared to the other groups. Consequently, PLGA coated β-TCP scaffold with sustained and localized release of VEGF showed favourable results for bone regeneration in vitro, and this scaffold has the potential to use as a drug delivery device in the future.

This study investigates a multifunctional hydrogel system integrating carboxymethyl cellulose (CMC) in a 3D-printed limonene (LIM) scaffold coated with poly(3,4-ethylenedioxythiophene): polystyrene sulfonate (PEDOT:PSS). The system allows to enhance wound healing, prevent infections, and monitor the healing progress. CMC is crosslinked with citric acid (CA) to form the hydrogel matrix (CMC-CA), while the 3D-printed limonene (LIM) scaffold is embedded within the hydrogel to provide mechanical support.

This study reports the discovery and preclinical activity of APS03118, a novel selective RET inhibitor featuring a novel tricyclic pyrazolo[3',4':3,4]pyrazolo[1,5-]pyridine hinge-binding scaffold designed to overcome acquired resistance to first-generation selective RET inhibitors (SRIs). By enhancing hydrogen bonding with conserved hinge residues (Glu805, Ala807), APS03118 potently inhibits wild-type RET and diverse resistance mutations, including solvent-front (G810R/S/C), gatekeeper (V804M/L/E), roof (L730I/M), and hinge (Y806C/N/H) variants. In preclinical models, APS03118 induced complete tumor regression in KIF5B-RET and CCDC6-RET V804 M patient-derived xenografts (PDXs) and significantly prolonged survival in an intracranial CCDC6-RET metastasis model.

Aims: This study aimed to enhance the osteoinductive potential of mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) by integrating them into a nano-hydroxyapatite (nHAp)-enriched hydrogel scaffold for bone regeneration applications.
Materials & Methods: EVs were isolated from naïve and osteogenically primed MSCs and characterized for morphology, cargo content, and cytocompatibility. Their uptake and osteoinductive activity were assessed using MC3T3 cells within a 3D interpenetrating network (IPN) hydrogel.

Hyperglycemia-induced oxidative stress and inflammation critically impair diabetic bone defect repair. Here, a radially oriented microchannel scaffold (D-GSH@QZ) was developed via a directional freezing technique integrated with photo-cross-linking strategies. The scaffold was fabricated from gelatin methacryloyl, silk fibroin methacryloyl, and nanohydroxyapatite (HAp) to mimic the natural bone matrix, while incorporating quercetin-loaded ZIF-8 nanoparticles (Qu@ZIF-8) for pathological microenvironment modulation.
