Article Abstract

We demonstrate a two-photon optogenetic method that generates action potentials in neurons with single-cell precision, using the red-shifted opsin C1V1(T). We applied the method to optically map synaptic circuits in mouse neocortical brain slices and to activate small dendritic regions and individual spines. Using a spatial light modulator, we split the laser beam onto several neurons and performed simultaneous optogenetic activation of selected neurons in three dimensions.

Adolescence is a sensitive period for frontal cortical development and cognitive maturation, marked by heightened structural plasticity in the dopaminergic (DA) mesofrontal circuit. However, the cellular and molecular mechanisms underlying this plasticity remain unclear. Here, we show that microglia, the brain's innate immune cells, are highly responsive to mesofrontal DA signaling during adolescence.

Two-photon holographic optogenetics has emerged as a powerful tool for achieving this via flexible excitation of user-defined neural ensembles. However, the precision of two-photon optogenetics has been constrained by off-target stimulation, an effect where proximal non-target neurons can be unintentionally activated due to imperfect spatial confinement of light onto target neurons.
